气相色谱/燃烧炉/同位素比质谱方法检测尿液中睾酮的来源

【摘要】 目的:建立检测尿液中睾酮来源的气相色谱/燃烧炉/同位素比质谱方法。方法:尿液经萃取等前处理后,采用HPLC方法分离尿液中睾酮及其代谢产物和激素前体PD,收集流出组分;流出组分经气相色谱/燃烧炉/同位素比质谱方法测定睾酮及其代谢产物和PD的δ13C值。结果:阴性尿液样品中睾酮及其代谢产物的δ13C值与PD的δ13C值的差值小于3。阳性尿液样品中睾酮及其代谢产物的δ13C值与PD的δ13C值的差值大于3。结论:该方法可以用于检测尿液中睾酮的来源。

【Abstract】 Aim: to develop a Gas Chromatography/Combustion/isotope-ratio mass spectrometry method to determine the origin of testosterone in urine. Methods: testosterone and metabolites in urine were extracted and purified using HPLC. The HPLC fractions were collected according to the retention time. The δ13C value of testosterone and metabolites in the fractions were analyzed by Gas Chromatography/Combustion/ isotope-ratio mass spectrometry. Result: the δ13C value measured for testosterone and metabolites in negative urine sample differs by less 3 delta units from that of the PD. the δ13C value measured for testosterone and metabolites in positive urine sample differs by over 3 delta units from that of the PD. Conclusion: The origin of testosterone in urine could be determined by measuring the δ13C values with this method.
