高效液相色谱法测定剑麻皂素提取液中剑麻皂甙元的含量

【摘要】 建立高效液相色谱测定剑麻皂素提取液中剑麻皂甙元含量的方法。色谱条件如下:采用RID-10A示差折光检测器,色谱柱填料为Hypersil ODS2 5μm,柱温为30℃,流速为l.0 m L/min,以乙醇-水(3+1,v/v)为流动相。实验表明,剑麻皂甙元在0.1~10 g·L-1范围内呈现良好的线性关系,线性相关系数为0.9998,RSD为0.32%(n=6)。在测定溶液中加入标准品做回收实验,测得回收率为98.38%。该检测方法简便快捷、精确稳定、重现性好,可为剑麻皂素生产提供实时监控和优化改进。

【Abstract】 A method for the determination of tigogenin in extract solutions by HPLC with RID was established. The chromatographic condition included: RID-10 A differential refractive index detector, The filler is Hypersil ODS2 5 μm was used as stationary phase, the column temperature at 30 ℃, the flow rate at l.0 m L/min and ethanol-water(3+1, v/v) as mobile phase. Linear relationship between values of peaks area and mass concentration was kept in the range of 0.1~10 g·L-1, with linear correlation coefficient of 0.9998, and RSD of 0.32 %(n=6). Standard was added in the determination of the recovery, the recovery rate was 98.38 %. The method of detection is simple and quick, accurate and stable, with good reproducibility, which can provide real-time monitoring and optimization in sisal saponin production.
