小麦镉胁迫相关microRNA的差异表达
Differential Expression of Cadmium Stress Related microRNA in Wheat
【摘要】 为探讨microRNAs(miRNAs)在小麦镉胁迫不同时间段的差异表达规律,初步鉴定参与小麦镉胁迫响应的miRNAs。以郑麦4号为材料,采用实时定量PCR技术研究小麦幼苗在不同时间镉胁迫(0h,6h,12h,24h,48h)后9个miRNAs的差异表达规律。结果表明:miRNA319和miRNA397在叶片和根中的表达呈不同程度的上调,而miRNA399和miRNA167在叶片和根中的表达呈不同程度的下调,暗示小麦幼苗中9个miRNAs参与了镉胁迫响应的调控。进一步对9个miRNAs作用的靶基因进行预测和分析发现,它们主要参与植物逆境条件下的信号传导、生长发育和胁迫应答等过程,可能在调控小麦对镉胁迫的反应中起着重要作用。
【Abstract】 In order to delve into the differential expression of microRNAs(miRNAs)in wheat treated with cadmium stress for different time,in this study,wheat seedlings of Zhengmai 4 were treated with different time(0,6h,12 h,24h,48h)under cadmium stress.Real-time PCR was used to detect the differential expression of nine miRNAs under cadmium stress.Results:Expression of miRNA319 and miRNA397increased in varying degrees in leaves and roots,while miRNA399 and miRNA167decreased,indicating that the nine miRNAs participated in regulation of cadmium stress response.The analysis on the target genes of these miRNAs revealed that these miRNAs participated in the signal transduction,growth and development and stress response under abiotic stress and might play important roles in wheat under cadmium stress.
