甘薯果糖-1,6-二磷酸醛缩酶5基因(IbFBA5)的克隆与生物信息学分析
Cloning and Bioinformatic Analysis of Fructose-1,6-Bisphosphate Aldolase 5 Gene in Sweetpotato(Ipomoea batatas L.)
【摘要】 本研究利用甘薯徐薯18和徐781转录组测序结果,筛选到一条与淀粉合成相关的差异表达Unigene,编号为T1-30656。以徐781为材料,克隆该片段,并对其进行重测序和生物信息学分析。结果表明,该片段为果糖-1,6-二磷酸醛缩酶基因的c DNA序列,该基因包含3个外显子和2个内含子,亚细胞定位在细胞质当中,将其命名为Ib FBA5。该c DNA长为1 341 bp,含1 074 bp的开放阅读框,编码357个氨基酸。其序列与番茄、油菜、粳稻和拟南芥等相似性在77%~79%之间。
【Abstract】 In this study we screened a differential expression unigene(sequencing number: T1-30656) from RNAseq data of Xushu18 and Xu781. The fragment was cloned from Xu781, and then it was sequenced and analyzed with bioinformatics approaches. Results showed that the cloned fragment encodes Fructose-1,6-Bisphosphate Aldolase(named Ib FBA5), containing 3 extrons and 2 introns, and it was subcellular localized in cytoplasm. The length of the c DNA is 1 341 bp with an open reading frame of 1 074 bp, which encodes 357 amino acid residues.The c DNA sequence is similar as these of Solanum lycopersicum, Brassica rapa, Oryza sativa Japonica Group and Arabidopsis thaliana, ranging from 77% to 79%.
