亚急性铜、镉暴露对鲤肝脏Hsp60表达的影响
Acute effect of copper and cadmium exposure on heat shock protein 60 in the liver of common carp(Cyprinuscarpio L.)
【摘要】 试验通过实时定量PCR和Western blot技术,检测重金属铜、镉单一及联合急性暴露后鲤肝脏中Hsp60基因和蛋白水平。鲤随机分为7组,两个Cu2+处理组(0.05 mg/l和0.1 mg/l),两个镉处理组(0.63 mg/l和1.26 mg/l),两个混合处理组(0.045 mg/l和0.09 mg/l)和对照组。结果表明铜、镉单一及联合急性暴露对鲤有毒性作用,经过96 h的暴露,除混合低浓度组之外,其余各组肝脏中Hsp60基因表达水平显著提高(P<0.05)。受重金属暴露的影响,初期鲤肝脏中Hsp60蛋白水平下降,随着时间推移,蛋白水平逐渐上升,混合低浓度组显著高于对照组水平(P<0.05)。
【Abstract】 In this paper, we investigated the m RNA and protein levels of HSP60 in the liver of commoncarp exposed to Cu, Cd and their mixture by western blot and real-time quantitative PCR in two Cu2+treatment groups(0.05 and 0.1 mg/l), two Cd2+treatment groups(0.63 and 1.26 mg/l), two mixture-treat-ment groups(0.045 and 0.09 mg/l) and one water control group The results indicated that single andcombined copper and cadmium exposure had toxic effects on common carp and significant increases(P<0.05)were observed in the Hsp60 m RNA levels in each exposure group except the low mixture group,after 96 h exposure. However, decreases of the Hsp60 protein level were observed in the beginning ofexposure. Subsequently, the Hsp60 protein level slowly increased over time. A significant increase wasobserved in the 1ow mixture exposure groups compared to the corresponding controls(P<0.05).
