姜黄素亚型B14对HKⅡ细胞损伤的保护作用及机制

【摘要】 采用CCK-8比色法检测了姜黄素(Cur)及其亚型对人肾皮质近曲小管上皮细胞(HKⅡ)的损伤作用;采用ELISA法检测了不同浓度和时间条件下血管紧张素Ⅱ(AngiotensinⅡ,AngⅡ)对HKⅡ细胞Cleaved caspase-3表达的影响,并建立了糖尿病肾病细胞模型;利用此模型研究了B14对HKⅡ细胞的保护作用及机制.结果表明:Cur及其亚型B13,B14对HKⅡ细胞均无毒害作用;以AngⅡ250nmol/L培养48h时,可建立最佳糖尿病肾病模型.B14对AngⅡ引起的HKⅡ细胞损伤具有保护作用,且此种保护作用与PI3K信号通路有关.

【Abstract】 The cell viability was detected with CCK-8 assay to observe toxic effects of curcumin(Cur)and its subtypes B13 or B14on HK Ⅱ.Then,cleaved caspase-3expression was detected with ELISA kit at different time and concentration of Angiotensin Ⅱ(Ang Ⅱ)in order to establish diabetic nephropathy cell model.Finally,the protective role and mechanism of Cur subtype B14 in the HK Ⅱwere researched with the cell model.The results showed that Cur and its subtypes B13 or B14had no toxic effects on the HK Ⅱ cells.Diabetic nephropathy cell model could be established with 250nmol/L Ang Ⅱfor 48 h.B14can protect HK Ⅱinjury induced by AngⅡand this process could be associated with PI3 K signaling pathway.This paper laid a foundation for researching the pathogenesis and developing new drugs of diabetic nephropathy.
