三七总皂苷对大鼠大脑缺血再灌注VEGF表达的影响
Effects of Panax Notoginseng Saponin on the Expression of Vascular Endothelial Growth Factor after the Brain Ischemia-reperfusion Injury in Rats
【摘要】 目的:观察三七总皂苷(PNS)对预处理大鼠大脑缺血再灌注后血管内皮细胞生长因子(VEGF)表达的影响。方法:将48只SD大鼠随机平均分为假手术组、模型组、PNS组和尼莫地平对照组。预给大鼠PNS 7d后,制备大鼠局灶性脑缺血再灌注模型。观察各组大鼠神经功能评分及大脑TTC染色,从VEGF的角度检测免疫组化和mRNA水平表达情况。结果:PNS可以显著改善大鼠神经功能评分和降低脑梗死体积,并可增加VEGF蛋白表达和上调VEGF mRNA表达水平。结论:PNS能有效上调脑缺血再灌注损伤后VEGF mRNA浓度的表达,促进缺血区血管新生。
【Abstract】 Objective:To observe effects of Panax Notoginseng Saponin(PSN)on the expression of Vascular Endothelial Growth Factor(VEGF)after the brain ischemia-reperfusion injury in rats.Methods:48SD rats had been randomly divided into 4groups:the sham operation group,the model group,Panax Notoginseng Saponin(PNS)group and Nimodipine group(n=12).The rats had been treated with PNS,and 7days later the rat focal cerebral ischemia-reperfusion models had been prepared.Neurobehavioral scores(NBS)had been evaluated in each group,TTC staining observed;the immunohistochemistry was used to observe VEGF and mRNA expressions.Results:PNS could not only improve significantly neurobehavioral scores and decrease dramatically cerebral infarct volume,but also increase remarkably VEGF and mRNA expression levels.Conclusion:The PNS is beneficial for rehabilitation after cerebral ischemia reperfusion injury via effectively up-regulating the injured cortical VEGF mRNA expression concentrations,which promotes vascular reborn in the ischemic region.
