雷公藤多苷片对糖尿病肾病大鼠肾组织WT1的影响

【摘要】 目的:探讨雷公藤多苷片(TW)对糖尿病肾病(DN)模型大鼠肾组织中足细胞特异性标志基因肾母细胞瘤抑制基因(WT1)表达的影响。方法:选用清洁级雄性SD大鼠30只,适应性喂养1周,随机分为3组:空白组、模型组、TW组,建立DN大鼠模型,TW灌服DN模型大鼠,6周后生化检测实验大鼠24h尿蛋白定量、血肌酐(Scr)、血尿素氮(BUN)、血浆白蛋白(ALB)、谷丙转氨酶(ALT);测量肾小球直径、肾小管短径、完整肾小管数目,并称取各组大鼠左肾质量、计算肥大指数;过碘酸-雪夫氏(PAS)染色观察肾脏病理;免疫组化检测肾组织WT1的表达。结果:TW能够上调实验大鼠肾组织WT1的表达,缩小肾小球直径及肾小管短径,减少肾质量及肥大指数,升高ALB,降低24h尿蛋白,与模型组比较差异显著(P<0.05,P<0.01),ALT组间比较差异无统计学意义。结论:TW可能通过影响DN大鼠肾组织WT1的表达,而改善上述生化指标,减轻肾脏病理损害。

【Abstract】 Objective: To investigate the effect of triperygium wilfordii multiglucoside tablets(TW) on foot cell specific protein marker Wilms’ tumor 1 gene(WT1) expression in rat renal tissue with diabetic nephropathy(DN). Methods: After adaptive feeding for a week, thirty male SD rats in clean grade were randomly divided into 3 groups: blank group, model group and TW group. DN rat model was established. TW group was given the TW by gavage. Some indicators, such as the quantity of 24 h urinary protein, serum creatinine(Scr), urea nitrogen(BUN), serum albumin(ALB) and alanine aminotransferase(ALT), were determined by means of biochemical analysis six weeks later. The glomerular diameter and renal tubular short diameter were measures and the number of complete renal tubules was counted. The weight of rat left renal was examined to calculate the hypertrophy index. Renal pathology was observed by periodic acid-schif s reaction(PAS); The expression of WT1 in renal tissue was examined by immunohistochemistry. Results: TW could up-regulate the expression of WT1 and increase the level of ALB. TW could shorten the glomerular diameter and renal tubular short diameter. And, TM could reduce the weight of kidney and the hypertrophy index. There was significant difference in the 24 h urinary protein between TM group and model group(P<0.05, P<0.01). There was, however, no difference in the level of ALT between TM group and model group. Conclusion: TW could regulate the expression of WT1 in rat renal tissue with diabetic nephropathy, which is likely to improve some biochemical indicators related to renal pathological damage.
