脐疗法治疗腹泻大鼠的血液和粪便代谢组学研究
Metabolomics research on the blood and faeces of diarrheal rats treated by umbilical compress therapy
【摘要】 目的:运用代谢组学方法研究小儿腹泻外敷散敷脐治疗大鼠腹泻的机制。方法:通过超高压液相色谱-四级杆飞行时间串联质谱(UPLC/Q-TOF-MS)分析药物作用下的大鼠血浆和粪便代谢物谱;使用主成分分析法(PCA)和正交偏最小二乘判别分析法(PLS-DA)研究各组之间的代谢物谱差异;采用变量重要性投影(VIP)确定标志物,通过对标志物浓度差异及代谢通路的研究,探讨药物对腹泻大鼠的干预效果。结果:初步确定了11个生物标记物。药物作用后,大鼠机体能量代谢、信号传导相关通路、肠道菌群代谢都受到不同程度的影响。结论:采用代谢组学的方法,较全面的反应了小儿腹泻外敷散敷脐治疗腹泻大鼠后的生理及代谢状态,为使用系统生物学研究敷脐疗法提供了依据。
【Abstract】 Objective: To investigate the mechanism of treating diarrheal rats with umbilical compressing with Xiaoer Fuxie Waifu Powder. Methods: UPLC/Q-TOF-MS was used to analyze the metabolites of plasma and feces of diarrheal rats treated by umbilical compress therapy. The principal component analysis(PCA) and orthogonal partial least-squares discriminant analysis(PLS-DA) were used to analyze the difference of metabolites among each group. Variable importance in projection(VIP) was used to identify the biomarkers. Investigate the intervention effect of drugs on diarrheal rats through researches on the concentration difference of markers and metabolic pathway. Results: 11 biomarkers were confirmed preliminary. The body’s energy metabolism, related signal transduction pathways and intestinal flora metabolism of diarrheal rats were influenced to varying degrees after treating with Xiaoer Fuxie Waifu Powder. Conclusion: Metabonomics method reflected the physiological and metabolic state of diarrheal rats after treating with Xiaoer Fuxie Waifu Powder, and provided a foundation for study on umbilical compress therapy with systems biology methods.
