宣肺平喘方对慢性哮喘大鼠肺组织TGF-β1、Smad2、Smad7蛋白表达的影响

【摘要】 目的:探讨转化生长因子(TGF-β1)、Smad2、Smad7在慢性哮喘大鼠肺组织中的定位表达情况,以反应其在哮喘大鼠气道重建过程中的作用。方法:将SD大鼠随机分为正常组、模型组、宣肺平喘方组和地塞米松组4组,每组10只。除正常组外,其余3组均利用卵蛋白雾化吸入诱发哮喘,宣肺平喘方组和地塞米松组在雾化吸入的同时给予宣肺平喘方及地塞米松治疗。肺组织病理切片(HE染色法),荧光免疫组化法测定TGF-β1、Smad2、Smad7蛋白在肺组织中的定位。结果:HE染色显示模型组支气管壁塌陷,周围有大量炎性细胞渗出;荧光免疫组化法显示TGF-β1、Smad2、Smad7蛋白均在支气管壁和肺泡壁上表达;模型组TGF-β1、Smad2蛋白含量明显高于正常组,治疗组与模型组相比明显下降(P<0.05);模型组Smad7蛋白含量明显低于正常组,治疗组与模型组相比明显上升(P<0.05)。结论:宣肺平喘方对哮喘气道重建有良好的治疗作用,能调节TGF-β1、Smad2、Smad7蛋白的水平。

【Abstract】 Objective: To explore the location expression of TGF-β1,smad2,smad7 in lung tissue of chronic asthma rats,and to reflect its effects in airway reconstruction of asthma rats.Methods: The SD rats were randomly divided into four groups: normal group,model group,Xuanfei Pingchuan Decoction group,dexamethasone group,with 10 rats in each group.Except for the normal group,chronic asthma model was achieved in all groups by inhaling ovalbumin.At the same time,Xuanfei Pingchuan Decoction group and dexamethasone group were given Xuanfei Pingchuan Decoction and dexamethasone respectively.The lung tissues biopsy(HE staining) and fluorescent immunohistochemical method were used to determine the position of TGF-β1,Smad2,Smad7 in lung tissues.Results: HE staining showed that there were a large number of inflammatory cells in the model group,the bronchus was collapsed.Immunofluorescence showed that TGF-β1,Smad2,Smad7 protein were located in the bronchial wall or alveolar wall.The content of TGF-β1 and Smad2 protein in the model group were significantly higher than these in normal group,while the treatment groups were decreased significantly than these in model group(P<0.05).The content of Smad7 protein in model group were significantly lower than these in normal group,while the treatment groups were increased significantly than these in model group(P<0.05).Conclusion: Xuanfei Pingchuan Decoction had a good effect on airway reconstruction of asthma,which could regulate the level of TGF-β1,Smad2,Smad7 proteins.
