沙苑子总黄酮对高脂大鼠的降甘油三酯作用及对肝脏DGAT2与ATGL的影响
Effects of complanatus flavonoids on triglyceride, DGAT2 and ATGL of hepatic in hyperlipidemia rats
【摘要】 目的:探讨沙苑子总黄酮对实验性高脂血症大鼠血脂及肝脏甘油三酯的影响及作用机制。方法:采用SD大鼠喂养高脂饲料的方法建立高脂血症大鼠模型。大鼠分为空白组、模型组、阳性药非诺贝特组、沙苑子总黄酮高、中、低剂量组,空白组和模型组给予0.9%氯化钠溶液,其余各组分别给予非诺贝特(100mg/kg)、沙苑子总黄酮(136、68、34mg/kg),连续灌胃4周。结果:沙苑子总黄酮高、中剂量可以显著降低血清甘油三酯水平(TG)、胆固醇水平(TC)和低密度脂蛋白胆固醇(LDH-C)水平(P<0.05);沙苑子总黄酮各剂量组均可显著降低肝脏TG、TC含量(P<0.05)。沙苑子总黄酮各剂量组均可下调肝脏DGAT2表达(P<0.05),而对大鼠肝脏ATGL表达没有显著影响。结论:沙苑子总黄酮可以调节高脂血症大鼠血清及肝脏脂质水平,沙苑子可通过下调肝脏DGAT2的表达而减少肝脏TG的合成,从而达到降低TG的作用。
【Abstract】 Objective: To study the effects of Complanatus fl avonoids on the blood lipids and triglyceride of hepatic in hyperlipidemia rats and effect mechanism. Method: SD male rats were fed with high fat diet for 8 weeks to establish the hyperlipidemia model. The rats were randomly divided into six groups according to the content of serum triglyceride and serum cholesterol: blank group, model group, positive(fenoi brate) group, and high dose, medium dose and low dose group of Complanatus fl avonoids. The control group and model group were gavaged with saline, the other groups were gavaged with fenoi brate(100mg/kg) and Complanatus extract(136, 68, 34mg/kg) for 4 weeks. Results: High and medium doses of Complanatus extract could significantly reduce serum triglyceride levels, cholesterol levels and LDL cholesterol levels(P<0.05), increase high-density lipoprotein levels(P<0.05). The contents of hepatic triglyceride and cholesterol were decreased in Complanatus fl avonoids groups(P<0.05) and the expression of DGAT2 was decreased(P<0.05). There was no significant effect on the expression of ATGL in rat liver. Conclusion: Complanatus fl avonoids could regulate the levels of serum and liver lipids in hyperlipidemic rats, down-regulate the expression of DGAT2 and reduce the triglyceride synthesis in liver.
