固本止咳中药对COPD模型小鼠脾、胸腺指数及Th1/Th2失衡的影响
Effects of Guben Zhike Recipe on the spleen index, thymus index and Th1/Th2 unbalance in COPD mice
【摘要】 目的:观察固本止咳中药对COPD模型小鼠脾、胸腺指数及Th1/Th2失衡的影响,探讨其治疗COPD的作用机制。方法:采用鼻腔滴入LPS加熏香烟的方法建立COPD小鼠模型,实验分为空白对照组、模型对照组、固本止咳中药组;运用HE染色对形态学进行观察,评价COPD小鼠模型;采用双抗体夹心ELISA方法检测小鼠血清中IFN-γ与IL-4分泌水平。结果:COPD小鼠模型形态学结果符合COPD临床病理形态学改变;模型对照组胸腺指数和脾脏指数显著低于空白对照组(P<0.05),与模型对照组比较,固本止咳中药组小鼠的胸腺指数和脾脏指数显著增加,差异有统计学意义(P<0.05)。与空白对照组比较,模型对照组小鼠血清中IFN-γ升高(P<0.05),IL-4降低(P<0.05),与模型对照组比较,固本止咳中药组小鼠血清中IFN-γ明显降低,IL-4升高,差异有统计学意义(P<0.05)。结论:固本止咳中药能纠正COPD模型小鼠Th1/Th2失衡,改善模型小鼠的免疫功能。
【Abstract】 Objective: To observe the effects and mechanism of Guben Zhike Recipe on the spleen index, thymus index and Th1/Th2 unbalance in COPD mice. Methods: The COPD model mice were established by cigarette smoke exposure and intratracheal instillation of lipopolysaccharide(LPS). The mice were divided into blank control group, model group and Guben Zhike Recipe group. The morphology characteristics were observed by H&E staining, to evaluate the COPD model mice. ELISA was used to detect the expression of interferon-γ(IFN-γ) and interleukin-4(IL-4). Results: The morphological results of COPD model accorded with the pathological characteristic of COPD in clinical. The thymus index and spleen index in model group was lower than that in the normal control group(P<0.05). And the thymus index and spleen index in Guben Zhike Recipe group was increased, the difference was statistically significance compared with the model group(P<0.05). The serum level of IFN-γ increased and the level of IL-4 decreased significantly in the model group(P<0.05), while the serum level of IFN-γ decreased and the level of IL-4 increased significantly in Guben Zhike Recipe group(P<0.05). Compared with the model group, the difference was statistically significance(P<0.05). Conclusion: Guben Zhike Recipe can correct the imbalance in Th1/Th2 and improve the immune function of COPD model mice.
