褪黑素保护红细胞抗过氧化氢氧化损伤作用研究
The protective effects of Melatonin on the anti-oxidative damage of RBCs
【摘要】 目的观察褪黑素(MT)对过氧化氢(H2O2)造成红细胞氧化损伤过程中的保护作用。方法将5 m L制备好的正常红细胞悬液分为对照组(0.9%Na Cl)、MT组(0.1 mmo L/L MT)、MT+H2O2组(0.1 mmo L/L MT+0.1mmo L/L H2O2)、H2O2+MT组(0.1 mmo L/L H2O2+0.1 mmo L/L MT)和H2O2组(0.1 mmo L/L H2O2),每组3份(红细胞悬液300μL/份),分别用H2O2和/或MT处理1 h后,通过分光光度法、硫代巴比妥酸(TBA)法及红细胞血红蛋白再释放试验(HRT)比较MT对H2O2氧化损伤红细胞后溶血率、上清液中丙二醛(MDA)含量及Hb再释放量的改变。结果对照组、MT组、MT+H2O2组、H2O2+MT组和H2O2组MDA含量(mmol/m L)分别为:1.19±0.04、1.24±0.08、1.41±0.06、3.18±0.05、3.73±0.07,与对照组相比,H2O2组、H2O2+MT组MDA含量明显增高(P<0.05);再释放条带灰度值分别为:176.55±7.98、171.23±11.23、184.27±9.36、230.17±8.98、239.22±10.47,与对照组、MT组和MT+H2O2组相比,H2O2+MT组和H2O2组再释放明显增高(P<0.05);溶血率分别为:0.061±0.007、0.070±0.005、0.135±0.007、0.145±0.011、0.117±0.015,与对照组和MT组相比,H2O2组、H2O2+MT组及MT+H2O2组溶血率明显增加(P<0.05)。结论提前加入褪黑素对H2O2造成的红细胞氧化损伤具有一定的保护作用,但是如果氧化损伤已经发生,则褪黑素对氧化损伤不再有明显的保护作用。
【Abstract】 Objective To observe the protective effect of Melatonin( MT) on the anti-oxidative damage of red blood cells( RBCs). Methods Five milliliters of prepared normal RBC suspension was divided into control( 0. 9% Na Cl),MT( 0. 1 mmo L/L MT),MT + H2O2( 0. 1 mmo L/L MT + 0. 1 mmo L/L H2O2),H2O2+ MT( 0. 1 mmo L / L H2O2+ 0. 1mmo L / L MT) and H2O2( 0. 1 mmo L/L H2O2) groups. Each group had 3 samples,and each sample had 300 μL of RBC suspension. After treating respectively with 0. 1mmol / L H2O2 and / or 0. 1mmol / L MT for 1 hour,hemolysis of RBCs,MDA and re-released hemoglobin( Hb) were detected by spectrophotometry,thiobarbituric acid method( TBA) and Hb re-release test( HRT). Results The MDA content( mmol / L) of control,MT,MT + H2O2,H2O2+ MT and H2O2 groups were1. 19 ± 0. 04,1. 24 ± 0. 08,1. 41 ± 0. 06,3. 18 ± 0. 05,and 3. 73 ± 0. 07 respectively. Compared with control,the MDA content of H2O2 and H2O2+ MT groups increased significantly( P < 0. 05). The gray values of re-released hemoglobin band of the groups above were 176. 55 ± 7. 98,171. 23 ± 11. 23,184. 27 ± 9. 36,230. 17 ± 8. 98,and 239. 22 ± 10. 47 respectively. Compared with control,MT and MT + H2O2 groups,the gray values of re-released hemoglobin band of H2O2+ MT and H2O2 groups increased significantly( P < 0. 05). The rates of hemolysis of RBCs of the groups above were 0. 061 ± 0. 007,0. 070 ± 0. 005,0. 135 ± 0. 007,0. 145 ± 0. 011,and 0. 117 ± 0. 015,respectively. Compared with control and MT group,MT + H2O2,H2O2+ MT and H2O2 group showed a significant increase in rates of hemolysis( P < 0. 05). Conclusion Pretreating RBCs with MT can protect the erythrocytes from oxidative injury caused by H2O2,but if the oxidative damage has already occurred,MT cannot protect RBCs from oxidative damage.
