全氟辛酸对小鼠睾丸生殖细胞线粒体过氧化损伤的研究
Study of Perfluorooctanoic Acid(PFOA) on Oxidative Damage in Mitochondria of Testicular Cells in Male Mice
【摘要】 为了研究全氟辛酸对雄性小鼠睾丸的氧化损伤,将雄性小鼠随机分为5组。连续对小鼠染毒14 d,用试剂盒法测定睾丸组织匀浆中还原型谷胱甘肽(GSH)、丙二醛(MDA)、琥珀酸脱氢酶(SDH)、超氧化物歧化酶(SOD)、乳酸脱氢酶(LDH)和ATP酶(Na+-K+-ATPase)的活性或含量的变化。研究结果表明:随着染毒量的增加,SOD的含量降低,MDA的含量升高,GSH的含量降低,SDH的活性则随着染毒量的增加而降低,ATPase的活性随染毒量的增加而明显降低,LDH的活性也随染毒量的增加而明显降低,因此全氟辛酸通过损伤睾丸细胞的线粒体,破坏其能量代谢,从而对睾丸组织产生损伤,导致睾丸坏死。
【Abstract】 To investigate the effects of toxicity of testicular in male mice poisoned by perfluorooctanoic acid,male mice were divided randomly into 5 groups in this experiment. After exposure for 14 d,superoxide dismutase(SOD),glutathione(GSH),malondialdehyde(MDA),succinate dehydrogenase(SDH),lactate dehydrogenase(LDH),and ATP enzyme(Na+-K+-ATPase) were measured by the method of reagent’s directions. The results show that activity of SOD in the model group decreased in a dose-dependent way; activity of MDA increased in a dose-dependent way; activity of GSH decreased in a dose-dependent way; activity of ATP enzyme decreased in a dose-dependent way; and activity of LDH decreased in a dose-dependent way. Therefore,perfluorooctanoic acid induced testicle by damaging mitochondria and energy decompensation,resulting in damage to the testicular tissues,finally caused testicle necrosis.
