硫化氢信号系统在张应力下大鼠骨髓间充质干细胞成骨分化过程中的变化

【摘要】 目的探讨张应力作用下大鼠骨髓间充质干细胞(BMMSCs)成骨向分化过程中硫化氢(H2S)信号系统的表达改变。方法应用四点弯曲加力系统,对体外分离培养的大鼠BMMSCs施加不同大小的周期单一张应力40min,2h后检测检测H2S信号系统中重要的H2S和胱硫醚-γ-裂解酶的表达量,同时检测碱性磷酸酶、骨钙素表达量并进行成骨细胞转录因子-2mRNA定量分析。结果随着张应力的增大,H2S信号系统的表达逐渐增强,同时成骨能力亦增强,但2000μstrain以后H2S信号系统的表达与成骨能力均下降。结论适当的张应力可促进H2S信号系统表达和BMMSCs骨向分化,硫化氢信号系统可能在这一过程中起重要作用。

【Abstract】 Objective To evaluate the changes of hydrogen sulfide signal system during the process of rat bone marrow-derived mesenchymal stem cells( BMMSCs) osteogenesis under tensile stress. Methods Different tensile stress was applied on rat BMMSCs by four-point bending apparatus for 40 minutes. Two hours later,the hydrogen sulfide and cystathionine-gamma-lyase were detected. Meanwhile,the changes of the alkaline phosphate,osteocalcin and runt-related transcription factor 2 mRNA level were also examined to evaluate the osteogenic ability. Results With the increase of tensile stress,the expression of H2 S signaling system gradually increased,so was the osteogenic capacity. However,both of the H2 S signal system expression and osteogenic capacity decreased obviously after the tensile stress exceeded 2000μstrain.Conclusion The proper tensile stress could promote hydrogen sulfide signal system expression and BMMSCs osteogensis.The hydrogen sulfide signal system may play an important role during this osteogensis process.
