响应面法优化枯草芽孢杆菌NS178产芽孢发酵工艺

【摘要】 应用单因素设计法对影响内生枯草芽孢杆菌NS178发酵液菌落数的因子进行筛选,得到3个主要影响因子,分别为葡萄糖、(NH4)2SO4、K2HPO4。采用Box-Behnken Design法对发酵培养基进行优化,通过响应面法分析获得最佳配方为葡萄糖2.29%、(NH4)2SO40.3%、K2HPO40.23%。对温度、初始p H值、装液量3组培养条件进行优化,最佳条件为33°C、p H 7.6、50 m L。在最佳配方和培养条件下,NS178发酵液菌落数为34.50×108cfu/m L;培养72 h时芽孢率达到75.8%;优化后比优化前发酵液抑菌圈直径增加67%。

【Abstract】 Three factors mainly influencing the colony number of endophytic Bacillus subtillus NS178 in fermentation liquid were screened out by single-factor experiments,which were glucose,( NH4)2SO4and K2HPO4. The Box-Behnken Design was applied to optimize the fermentation medium for NS178,and the optimum one was obtained as 2. 29% glucose,0. 3%( NH4)2SO4and 0. 23% K2HPO4 after the analysis of response surface methodology. The culture conditions were also optimized,and the optimal conditions were temperature as 33℃,p H value as 7. 6 and fluid volume as 50 m L. Under the optimal fermentation conditions,the yield of NS178 was 34. 50 × 108 cfu / m L; the spore rate could reach 75. 8% after cultured for 72 hours; and the diameter of inhibition zone increased by 67% compared to that before fermentation.
