运用网络生物学方法分析双黄连口服液治疗甲型H1N1流感的药理机制
Analysis on Pharmacological Mechanism of Shuanghuanglian Oral Liquid for Influenza A (H1N1) by Network Biology Approaches
【摘要】 目的探讨双黄连口服液治疗甲型H1N1流感的药理机制。方法文本挖掘文献证实甲型H1N1流感相关的基因或蛋白质,查询PubChem数据库获得双黄连口服液的靶蛋白,依据多个相互作用数据库中数据,运用网络生物学方法分别构建甲型H1N1流感体内反应的蛋白质相互作用网络和双黄连口服液的靶蛋白相互作用网络,将后者映射到前者,建立双黄连口服液对抗甲型H1N1流感的靶蛋白相互作用网络,并进行网络拓扑结构和基因本位论分析。结果双黄连口服液药理作用主要与细胞对外部刺激反应的调节、细胞凋亡的调节和信号转导等有关,主要涉及糖原合成激酶-3B(GSK3B)、肿瘤基因P53(TP53)、核因子-κBp50(NFKB1)、热休克蛋白90-alpha(HSP90AA1)等。结论双黄连口服液可能主要是通过影响GSK3B等信号通路,调节细胞凋亡而减少病毒复制,抑制甲型H1N1流感的疾病进程。
【Abstract】 Objective To study the pharmacological mechanism of Shuanghuanglian Oral Liquid( 双黄连口服 液) for influenza A( H1N1). Methods Genes or proteins related to influenza A( H1N1) were obtained by text mining,and target proteins of Shuanghuanglian Oral Liquid. were searched in PubChem. Influenza A( H1N1) network and Shuanghuanglian Oral Liquid network were constructed by interaction between the genes or proteins related to influenza A( H1N1) and target proteins of Shuanghuanglian Oral Liquid from multiple protein-protein interaction databases respectively. The sub-network of Shuanghuanglian Oral Liquid for influenza A( H1N1) was produced by mapping Shuanghuanglian Oral Liquid network to influenza A( H1N1) network. The networks were analyzed with network topology and gene ontology. Results The pharmacological mechanism of Shuanghuanglian Oral Liquid was related to stimulus,regulation of apoptosis and signal transduction. Glycogen synthase kinase-3B( GSK3B),tumor gene P53( TP53),nuclear factor-κBp50( NFKB1),and heat shock protein 90-alpha( HSP90AA1) were involved. Conclusion Shuanghuanglian Oral Liquid can inhibit the disease progression of influenza A( H1N1) by affecting GSK3B signaling pathways,regulating apoptosis,and reducing viral replication.
