乌司他丁注射液改善急性肺损伤兔循环内皮祖细胞成血管能力及对PKB/eNOS/NO通路的影响

【摘要】 目的观察乌司他丁注射液对急性肺损伤循环内皮祖细胞血管生成能力及对PKB/eNOS/NO通路的影响。方法体外细胞实验共分为5组:正常对照组、急性肺损伤组、乌司他丁治疗组、乌司他丁+LY294002组,乌司他丁+L NAME组分别进行体外皮祖细胞成血管能力检测;PKB、eNOS蛋白检测;NO浓度检测。结果乌司他丁治疗组较急性肺损伤组体外成血管数目增加,PKB、eNOS蛋白表达上升,NO浓度上升,差异有统计学意义(P<0.05)。结论乌司他丁注射液通过PKB/eNOS/NO通路,提高循环内皮祖细胞成血管能力。
【Abstract】 Objective To observe the ability of angiogenesis in acute lung injury circulating endothelial progenitor cells after uli-nastatin injected and angiogenesis of PKB/eNOS/NO pathway affected.Methods The cells in vitro were divided into 5 groups:The control group,acute lung injury,ulinastatin treatment group,ulinastatin treatment + LY294002 group,ulinastatin treatment + L NAME groups.Angiogenicability,PKB,eNOS protein,NO concentration were detected.Results The ability of angiogenesis in ulinas-tatin treatment group was higher than that in acute lung injury groups(P <0.05).The PKB,eNOS protein expression increased in ulinastatin treatment group,and there was differences between ulinastatin treatment group and acute lung injury groups(P <0.05).NO concentration was increased in ulinastatin treatment group,and there was differences between ulinastatin treatment group and a-cute lung injury groups(P <0.05).Conclusion Ulinastatin injection could enhance the ability of angiogenicability on circulating en-dothelial progenitor cells through PKB/ eNOS /NO pathway.
