大黄庶虫虫丸对动脉粥样硬化大鼠血管保护作用的研究

【摘要】 目的:观察大黄庶虫动脉粥样硬化大鼠模型MDA、SOD、ET、NO及核因子-KB表达的影响,旨在从抗氧化损伤、内皮细胞功能、生物因子表达等各方面探讨保护血管的作用机制。方法:用高脂饲料喂养加vitD3建立大鼠AS模型,用大黄庶虫虫丸混悬液高(1.4g生药/kg)、低(0.7g生药/kg)剂量分别饲喂8周后,处死大鼠,观察各组大鼠丙二醛(MDA),超氧化物岐化酶(SOD),内皮素(ET)、一氧化氮(NO),同时采用免疫组化法观察NF-kappaB蛋白表达。结果:大黄庶虫虫丸高剂量组(1.4g生药/kg)与模型组对比NO显著升高,ET水平明显降低;大黄庶虫虫丸低剂量组(0.7g生药/kg)与模型组对比血清SOD活力升高,大黄庶虫虫丸高剂量组(1.4g生药/kg)与模型组对比血清SOD活力显著升高,MDA含量降低;大黄庶虫虫丸高低剂量组与模型组相比血管壁NF-kappaB蛋白表达明显减弱。结论:大黄庶虫虫丸通过改善内皮细胞功能,减轻氧化损伤,抑制动脉血管NF-kappaB的表达,起到保护血管抗动脉粥样硬化的作用。

【Abstract】 Objective: Observation of atherosclerosis in rats model of MDA,SOD,ET,NO and influence of nuclear factor- KB from oxidative damage resistance,endothelial cell function,biological factor expression and so on various aspects to study the effect of protecting vascular system. Methods: With high fat feed vitD3 AS rats model was established,using dahuang zhechong pill mixed suspension high crude drugs( 1.4 g/kg),low crude drugs( 0.7 g/kg) dose feeding 8 weeks after respectively,put to death in rats,observe each group rats malondialdehyde( MDA),superoxide disproportionation enzyme( SOD),endothelin( ET),nitric oxide( NO),at the same time using immunohistochemical method to observe the NF kappaB protein expression. Results: Dahuang zhechong pill crude drugs( 1. 4 g /kg),high dose group compared with model group,NO significant rise,ET level is decreased obviously( P < 0. 01,P < 0. 05); Dahuang zhechong pill crude drugs( 0.7 g/kg),low dose group compared with model group serum SOD activity increased( P <0.05),dahuang zhechong pill crude drugs( 1.4 g /kg),high dose group compared with model group significantly increased serum SOD activity( P < 0. 01),lower MDA content( P < 0.05); Dahuang zhechong pill high and low dose group and model group compared with the vessel wall NF kappaB- protein expression decreased significantly( P < 0. 01). Conclusion: dahuang zhechong pill by improving endothelial cell function,reduce oxidative damage and inhibit the expression of NF- kappaB artery,protect the vascular resistance to atherosclerosis.
