玉屏风散对变应性鼻炎肥大细胞活性的抑制作用

【摘要】 目的:从肥大细胞(Mast cells,MCs)的活性探讨玉屏风散对变应性鼻炎(AR)的作用及其作用机制。方法:将SD大鼠60只随机分为空白组、模型组、阳性药酮替芬组和玉屏风散12、6及3g/kg剂量组,每组10只;除空白组外,其余各组联合卵蛋白及氢氧化铝复制AR大鼠模型。给药组大鼠分别灌胃给予0.46g/kg酮替芬及12、6及3g/kg的玉屏风散,1次/天。实验结束,取鼻粘膜进行甲苯胺蓝染色,观察组织中MCs数量;采用体外细胞培养和血清药理学方法,观察玉屏风散含药血清对变应性鼻炎动物模型腹腔肥大细胞(RPMC)释放类胰蛋白酶的影响;蛋白印记检测RPMC释放类胰蛋白酶的表达水平。结果:体内实验表明:与空白组比较,模型组可见明显的肥大细胞脱颗粒现象,颗粒位于细胞之外,肥大细胞失去原有形态,难以辨认;玉屏风散12g/kg组和酮替芬组肥大细胞脱颗粒比模型组明显降低。体外实验显示:与模型组含药血清组比较,玉屏风散12g/kg血清组RPMC释放类胰蛋白酶的量明显降低(P<0.05,P<0.01);与空白组比较,模型组类胰蛋白酶表达水平明显升高,经玉屏风散治疗后表达降低。结论:玉屏风散12g/kg在体内可抑制变应性鼻炎MCs脱颗粒,体外可抑制变应性鼻炎RPMC脱颗粒释放类胰蛋白酶,并抑制类胰蛋白酶的表达,从而最终抑制变应性鼻炎肥大细胞的活性,以抑制其过敏症状。

【Abstract】 Objective:To Investigation on the action and mechanism of Yupingfeng powder to allergic rhinitis by observation of the activity of mast cells.Methods:60 rats were randomly divided into control group,model group,ketotifen fumarate group and three groups of Yupingfeng powder( 12,6,3g / kg).We copied the allergic rhinitis rats combing the egg albumin and Al( OH) 3 and administrated the model rats with Yupingfeng powder and ketotifen fumarate;observed the mast cells in the tissue of nasal mucosa stained by histological means named toluidine blue staining.Using the cell-culture in vitro and the method of serum pharmacology,we observe effect of serum containing Yupingfeng powder to the release of tryptase from the peritoneal mast cells( PRMCs).Using the Western blot and the method of serum pharmacology,we study the expression of tryptase from the PRMcs.Results:Administrated with Yupingfeng powder by 12g / kg,the model group showed significant degranulation of mast cells and the granules were outside the cells.The mast cells lose their original shape,and were difficult to identify;degranulation of mast cells in Yupingfeng powder group and ketotifen fumarate group was significantly lower than model group.In vitro,compared with the model group,the releasing of tryptase in RPMC happening in groups containing Yupingfeng serum was significantly reduced,and the difference was statistically inhibit the releasing of tryptase produced by degranulation of RPMC from allergic rhinitis tissues significant( P <0.05,P <0.01,P <0.001).The level of expression of tryptase increased greatly comparing to the control group and decreased remarkably after being administrated with Yupingfeng powder.Conclusion:In vivo and in vitro,Yupingfeng San can stabilize the cell membrane of mast cells,inhibit the degranulation of mast cells and prevent the release of tryptase from the mast cells,which could inhibit the allergy symptoms.
