【摘要】 目的分析苏素特异蛋白酶1(SENP-1)在脐带血造血干细胞(UCB-HSCs)中的表达情况。方法通过逆转录多聚酶连反应(RT-PCR)法检测SENP-1分子mRNA水平,以?-action为内对照;实验分为对照组A组、B组,实验组C组、D组、E组,每组检测15例标本。结果对照组中A组和B组均未表达SENP-1mRNA,实验组中C组、D组、E组均表达SENP-1类分子mRNA,以SENP-1mRNA积分光密度值/?-actin mRNA积分光密度值比较各组冻存前后及冻存时间长短SENP-1表达差异,C组为0.146±0.011,D组为0.314±0.011,E组为0.368±0.019。结论白血病细胞(HL-60)不表达SENP-1mRNA分子,即使经炎症因子r-干扰素(IFN-r)刺激也不表达,不能有效激活CD8+杀伤T细胞(CTL),是发生免疫逃逸的重要原因;脐带血造血干细胞中均有SENP-1的表达,随着冻存时间越久,SENP-1表达量越下降,表明冻存可以使脐带血干细胞的免疫原性下降。更多还原

【Abstract】 Objective:Study on the expression of SENP-1 in umbilical cord blood hematopoietic stem cells(UCB-HSCs).Methods:To examine the level of expression of SENP-1mRNA by RT-PCR.Useing β-action as internal control.The exprimental group was divided into A group and B group,The control group was divided into C group,D group and E group.Each group had 15 cases.Results:There was no expression of SENP-1mRNA in control group and there was obvious expression in exprimental group.Complare the different expressions of SENP-1 in every group by SENP-1mRNA/β-action.The C group was 0.146±0.011,the D group was 0.314±0.011,the E group was 0.368±0.019.Conclusion:The expression of SENP-1mRNA was not detectable in the leukemia cell HL-60,there was no expression even if by the stumilation of IFN-r,so it can’t effectively activate CTL cells,this was the important reason of immune escape;the expression of SENP-1mRNA was detectable in every exprimental group,the expression of SENP-1mRNA was decreased when the frozen storage time was increasing,this indicated that frozen storage can make immunogenicity of UCB-HSCs reduced.更多还原
