【摘要】 目的:观察12-去氧佛波醇13-棕榈酸酯(12-Deoxyphorbol-13-Palmitate,12D-13P)对慢性粒细胞白血病(chronic myeloid leukemia,CML)细胞株K562细胞增殖的抑制作用。方法:不同浓度的12D-13P处理的K562细胞,光镜下观察细胞形态学变化,MTT法检测药物对K562细胞增殖的抑制作用,甲基纤维素克隆形成实验分析细胞克隆能力。结果:MTT法和甲基纤维素克隆形成实验均显示12D-13P对K562细胞增殖具有较明显的抑制作用。结论:12D-13P能抑制体外培养的K562细胞的增殖,且呈一定的量效和时效关系。更多还原

【Abstract】 Objective: To observe the inhibitive effect of 12-Deoxyphorbol-13-Palmitate( 12D-13P) on the cell proliferation of chronic myeloid leukemia cell line K562. Methods: K562 cells were treated with different concentrations of 12D-13 P. Morphology changes were observed with optical microscope. Medicine role in cell proliferation was detected with MTT assay. Cell cloning ability was analyzed with methyl cellulose colony-forming experiment. Results: MTT methods and methyl cellulose colony-forming experiment indicated 12D-13 P had an obviously inhibitory effect on K562 cells. Conclusion: 12D-13 P can significantly inhibit the growth of in vitro cultured K562 cells and it shows a dose effect and time effect relationship.更多还原
