姜黄素对人皮肤鳞状细胞癌细胞SCL-1凋亡的影响及相关机制研究

【摘要】 目的:探讨不同浓度姜黄素对人皮肤鳞状细胞癌细胞系SCL-1增殖及凋亡的影响,并探讨其可能的作用机制。方法:以体外培养的人皮肤鳞癌细胞系SCL-1为研究对象,用不同浓度的姜黄素处理后,CCK-8法观察姜黄素对SCL-1细胞的生长抑制作用,AnnxinV-FITC/PI法检测细胞早期凋亡,实时荧光定量RT-PCR法检测Caspase-3、Caspase-9 mRNA的表达,酶标比色法检测Caspase-3及-9的活性。结果:浓度大于10μmol·L-1姜黄素能显著抑制SCL-1细胞的增殖,诱导其凋亡,并上调细胞内Caspase-3及-9 mRNA的表达及活性。结论:姜黄素能抑制SCL-1细胞的增殖、诱导其凋亡,上调Caspase-3及-9的表达及活性是其可能的分子机制之一。

【Abstract】 Objective: To investigate the effect of curcumin on the apoptosis of SCL-1 cell and its related mechanism. Methods: Human cutaneous squamous cell carcinoma cell line SCL-1 was treated with different concentrations of curcumin. The cell growth inhibition was evaluated by CCK-8 assay,and early apoptosis was assessed with AnnxinV-FITC /PI. The real- time quantitative RT- PCR was used to detect the mRNA expression of caspase-3 and Caspase-9,and the activity of caspase-3and caspase-9 mRNA were determined by enzyme standard colorimetric method. Results: Concentration of curcumin above 10 μmol·L- 1significantly inhibited the growth of SCL-1cells proliferation,and induced the apoptosis and increased the expression and activity of caspase-3 and Caspase-9. Conclusion: Curcumin could inhibit the growth of SCL-1 cells proliferation and induce apoptosis,which may be related to the upregulation of the expression and activity of caspase-3 and caspase-9.
