人参皂苷Rh2通过GATA-1诱导K562细胞红系分化作用的研究
Effect of Ginsenoside Rh2 on Inducing Erythroid Differentiation of K562 Cells through GATA-1
【摘要】 目的了解人参皂苷Rh2是否通过GATA-1蛋白诱导白血病细胞株K562的分化作用。方法利用凝胶集落生成实验检测不同浓度人参皂苷Rh2对白血病细胞株K562的增殖抑制作用;采用免疫细胞化学法和Western-blot法检测不同浓度人参皂苷Rh2处理前后K562细胞株中GATA-1蛋白表达情况。结果凝胶集落生成实验结果显示人参皂苷Rh2对K562细胞有明显的增殖抑制作用,且呈剂量依赖性;免疫细胞化学法和Western-blot法提示人参皂苷Rh2可诱导K562细胞内GATA-1表达,且呈剂量依赖性。结论人参皂苷Rh2可显著抑制白血病细胞株K562的生长,其作用呈浓度依赖性;人参皂苷Rh2在体外可通过上调GATA-1蛋白表达诱导K562向红系细胞方向分化。
【Abstract】 Objective: To investigate whether the ginsenoside Rh2 could induce the differentiation of K562 cells through GATA-1. Methods: K562 cells were treated with different concentrations of ginsenoside Rh2. Gel colony forming experiment was used to detect the proliferation. Immunocytochemical and Western-blot were used to detect the expression of GATA-1 in K562 cells. Results: Ginsenoside Rh2 could significantly inhibit proliferation and enhance expression of GATA-1 in K562. Conclusion: Ginsenoside Rh2 can significantly inhibit the growth of K562 and induce its erythroid differentiations.
