参麦注射液保护D-NNA所致大鼠肾脏氧化损伤的研究

【摘要】 目的:通过研究参麦注射液保护D-NNA所致大鼠肾脏损伤,揭示参麦注射液保护肾脏组织的机制。方法:建立D-NNA诱导大鼠致肾脏损伤模型,用参麦注射液治疗模型组大鼠,通过检测血清生化指标Crea和Bun;肾脏氧化指标MDA、GSH-Px和SOD;一氧化氮水平并结合病理组织学研究,分析D-NNA造模情况和参麦注射液治疗效果。结果:经D-NNA诱导的大鼠血清生化指标Crea和Bun水平均升高,肾脏组织中MDA浓度和GSH-Px活性明显升高,SOD活性明显下降;肾脏组织中NO水平降低,病理学检查证明有严重的肾脏病变。经参麦注射液治疗后所有指标均有改善,病理学检查显示肾脏组织有明显恢复。结论:参麦注射液具有保护D-NNA所致大鼠的肾脏损伤,各氧化指标、NO水平等指标证实其主要机制为参麦注射液在肾脏内具有抗氧化作用。

【Abstract】 Objective: To investigate the protection of Shenmai Injection for kidney damage induced by D- NNA and to reveal the mechanism of protecting kidney tissue by Shenmai Injection. Method: We constructed a model of kidney damage by intraperitoneally injecting D- NNA,and then Shenmai Injection was used. After that,we collected serum to detect the Crea and Bun levels. Kidney tissues were gathered for getting the MDA concentration and SOD,GSH- Px activities,NO level expression in kidney tissue would be examined,respectively. The entire index above combined with histopathology was used to evaluate the model of kidney damage induced by D- NNA and the cured effect of Shenmai Injection was assessed. Result: The rats,induced by D- NNA,showed significant rise of Bun,Crea and MDA levels and significant decrease of SOD,GSH- Px activities and NO level as compared with control group. In addition,the kidney of rats induced by D- NNA showed remarkable histopathology. After Shenmai Injection treatment,the entire index above trended to become normal,and pathological examination showed the kidney tissue partly recovered. Conclusion: Shenmai Injection can protect kidney damage induced by D- NNA,and the mechanism of protection is antioxidation. Meanwhile,all of the indexes studied for oxidation cooperates with histopathological exam tend to ensure this improvement.
