代谢型谷氨酸受体5mRNA与脑梗死的相关性研究
The Study on Relationship between Metabotropic Glutamate Receptor 5 mRNA and Cerebral Infarction
【摘要】 目的:观察急性脑缺血大鼠脑组织中mGluR5 mRNA在不同时段的表达变化的规律,探讨mGluR5mRNA与脑梗死的相关性。方法:75只雄性Wistar大鼠按照随机数字表法分为正常对照组、手术组1、3、6、12、24 h及3 d、14 d组及其相对应的假手术对照组,采用大鼠MCAO模型,利用原位杂交技术,检测各组mGluR5mRNA的表达。结果:与正常对照组相比,假手术对照组mGluR5 mRNA表达无明显改变(P>0.05)。手术组mGluR5 mRNA各时间段表达均显著高于假手术对照组及正常对照组,缺血1 h其表达即有升高,比较差异均有统计学意义(P<0.01)。mGluR5 mRNA表达6 h达到高峰(F=5.328,P<0.00),3 d后表达开始下降(P<0.01),14 d基本降至正常对照水平(P>0.05)。结论:mGluR5在脑缺血后升高,提示mGluR5在脑梗死中有着重要作用,从而对我们的临床研究指出一条途径。
【Abstract】 Objective:To investigate the correlation between mGluR 5mRNA and cerebral infarction.Method: Seventy-five male Wistar rats were randomly divided into normal control group, operation group 1, 3, 6, 12, 24 h and 3 d, 14 d groups and corresponding sham operation control group, the rat MCAO model was used in situ hybridization, was used to detect the expression of mGluR5 mRNA.Result:The normal group and the sham operation group had no difference at each time(P>0.05). Each time the operation group mGluR5 mRNA expression were higher than the sham operation control group and normal control group, ischemia 1 h and its expression is increased, and the difference were all statistically significant(P<0.05). The mGluR5 mRNA expression reached peak at 6 h(F=5.328, P<0.00), began to decline after 3 d(P<0.01), the basic dropped to normal control level after 14 d(P>0.05).Conclusion:MGluR5 increased after cerebral ischemia, suggesting that mGluR5 plays an important role in cerebral infarction, so as to point out a way for our clinical study.
