【摘要】 [目的]探讨Mel-18在结肠癌组织和正常肠黏膜组织中的表达及其与临床病理特征的关系,初步评价Mel-18在结肠癌发生、发展中的作用及意义。[方法]共收集48例结肠癌患者的癌组织及其对应的正常肠黏膜组织,采用RT-PCR方法检测结肠癌组织与对应正常肠黏膜组织Mel-18mRNA的表达情况,并分析其与临床病理指标的关系。[结果]Mel-18mRNA在结肠癌组织及对应正常肠黏膜组织中的相对表达量分别为(0.932±0.128)和(2.023±0.176),Mel-18mRNA在结肠癌组织中呈明显低表达,相对表达率为相应正常肠黏膜组织的0.48倍。Mel-18的表达与性别、年龄和分期之间没有明显的相关性。在有淋巴结转移的患者组中Mel-18的表达比没有转移组要低,差异无统计学意义。[结论]Mel-18在结肠癌中表达下降可能与结肠癌的发生有关,可能作为一种反映结肠癌生物学行为的有效指标。更多还原

【Abstract】 [Objective]To investigate the expression of Mel18in colorectal cancer tissue,normal colon mucosa tissues on RNA levels,and its relationship with clinicopathologic significance in colorectal cancer and to elucidate their impact on the carcinogenesis and progression of colorectal.[Methods]The cancer samples and normal colon mucosa samples from 48colon cancer patients were collected and the expression of Mel18mRNA was detected by reverse transcriptase polymerase chain reaction(RTPCR).Statistic alanalysis was performed to investigate the association between the clinicopathologic features and Mel18expression.[Results]The results of realtime PCR analysis showed the expression of Mel18mRNA was 0.932± 0.128and 2.023±0.176in colorectal cancer tissue and normal colon mucosa respectively.Significant downregulation for Mel18rnRNA was detected in colorectal cancer tissues.Relative expression abundance of Mel18mRNA in colorectal cancer tissues was 0.48times in normal colon mucosa.There was no correlation between Mel18expression and clinicopathological parameters,such as age,gender and clinical stage(P >0.05).The decrease of Mel18expression was negatively correlated with lymph node metastasis but there was no obvious statistical significance.[Conclusion]The abnormal expression of Mel18in colon carcinoma is correlated with its carcinogenesis,which may be a new effective prognostic factor to indicate biologic behavior of colon cancer.更多还原
