ADAMTS-1对小鼠肺成纤维细胞Ⅰ型胶原蛋白的影响
ADAMTS-1 influence expression of type 1 collagen in vitro
【摘要】 目的探讨含Ⅰ型血小板结合蛋白基序的解聚蛋白样金属蛋白酶(ADAMTS-1)对小鼠肺成纤维细胞Ⅰ型胶原蛋白(COL1)表达的影响及可能的机制。方法应用细胞培养技术进行小鼠肺成纤维细胞培养,利用ADAMTS-1为刺激因子,将培养的细胞分为空白组、低、中、高浓度组,应用逆转录-多聚酶链反应技术(RT-PCR)观察COL1 mRNA、转化生长因子β1(TGF-β1)mRNA在各组中的表达情况;采用蛋白质印迹法(Western blotting)观察各组细胞COL1、TGF-β1的变化。结果与空白组相比,低浓度组、中浓度组、高浓度组的COL1、TGF-β1表达水平下降,其中高浓度组表达水平最低(P<0.05)。结论 ADAMTS-1能降低COL1的表达,抑制TGF-β1是其抗纤维化作用的可能潜在机制。
【Abstract】 【Objective】 To explore the influence of ADAMTS- 1 on expression of collagen Ⅰ in vitro and its possible mechanism. 【Methods】 Mouse lung fibroblasts were cultured. They were assigned to fore groups: normal control group, low concentration group, medium concentration group, high concentration group. Type I collagen and TGF- β1were measured by RT-PCR and western blotting. 【Results】 Compared with normal control group, the levels of Type I collagen and TGF- β1were significantly decreased in other three groups, especially in high concentration group(P <0.05). 【Conclusion】 ADAMTS-1 can degrade Type I collagen, inhibiting TGF- β1may be a potential mechanism of its anti-fibrosis.
