左卡尼汀对动脉粥样硬化血管平滑肌细胞肉碱棕榈酰转移酶1表达的干预研究
Intervention study L-carnitine on carnitine palmitoyltransferase-1 expression of vascular smooth muscle cells in atherosclerosis
【摘要】 目的探讨左卡尼汀对动脉粥样硬化模型兔主动脉平滑肌细胞层肉碱棕榈酰转移酶1(CPT-1)的表达变化。方法 45只新西兰大白兔分成对照组、高脂组和干预组。高脂组与干预组予以高脂饲料(每单位质量含5%猪油及1%胆固醇),在此基础上,干预组给予左卡尼汀,100 mg/kg剂量肌肉注射每天1次。在饲养24周后,采用逆转录聚合酶链反应及免疫组化方法检测血管壁平滑肌细胞层CPT-1 mRNA及蛋白的表达水平。结果同对照组比较,高脂组及干预组CPT-1 mRNA及蛋白的表达明显减少,且高脂组减少更为明显(F=192.282、93.329,P均<0.05)。结论左卡尼汀可通过增加血管平滑肌CPT-1的表达从而起到抗动脉粥样硬化的作用。
【Abstract】 Objective To investigate the effect of L-carnitine on carnitine palmitoyltransferase-1(CPT-1) expression of vascular smooth muscle cells in atherosclerotic rabbits. Methods A total of 45 rabbits were randomly assigned to the control group, high-cholesterol diet group and treatment group, 15 rabbits in each group. The rabbits in the high-cholesterol diet group and treatment group were treated with high-fat diet(per unit mass containing 5% lard and 1%cholesterol), and the rabbits in the treatment group received L-carnitine by intramuscular injection(100 mg / kg, once daily) additionally. The expression of CPT-1 mRNA was detected by reverse transcription polymerase chain reaction and the CPT-1 protein was determined by immunohistochemistry after 24-week treatment. Results Compared with the control group, the expression of CPT-1 mRNA and protein in the high-cholesterol diet group and treatment group were much lower, and were lowest in the high-cholesterol diet group(F = 192.282, 93.329, all P <0.05). Conclusion L-carnitine can increase the expression of CPT-1 in vascular smooth muscle cells and play an important role in atherosclerosis.
