离子色谱法快速测定血清中甲基丙二酸含量

【摘要】 目的建立简便快速梯度淋洗离子色谱法测定血清中甲基丙二酸的含量。方法血清样品经超声提取,12 000 rpm/min高速离心后,再用On Guard II RP柱、On Guard II Ag柱和On Guard II Na柱净化除去杂质,最后用0.25μm滤膜过滤后直接进样。选用Ion Pac AS23阴离子分析柱(4 mm×250 mm),用KOH淋洗液(0 mmol/L~35 mmol/L)梯度淋洗,流速为1.0 ml/min,使用电导检测器进行检测。结果本法线性相关系数r=0.9995,精密度RSD<10%,样品加标平均回收率为88.8%~101%,最低检出量为0.0063μg/ml。结论成功建立了准确测定血清中甲基丙二酸浓度的方法,样本前处理简单、快速,方法准确灵敏,可以用于甲基丙二酸血症(MMA)的筛查、诊断和治疗效果的监测。

【Abstract】 Objective To establish a method of ion chromatographic with simple and fast gradient elution for the determination of methylmalonic acid in serum. Methods The serum samples were firstly extracted under ultrasound,and then centrifugal at12 000 r / min,and thirdly cleaning by On Guard II RP( 1. 0 ml),On Guard II Ag,and On Guard II Na SPE column,and finally filtering by 0. 25 μm membrane. The separation was carried on an Ion Pac AS23 analytical column with a gradient eluent of0 mmol / L ~ 35 mmol / L KOH and at a flow rate of 1. 0 ml / min and detected by suppressed conductivity detector. Results The method had a good linear correlation( r) 0. 9995 and precision( RSD < 10%). The average recoveries were 88. 8% ~ 101%and the detection limit was 0. 0063 μg / ml. Conclusion The method is simple,quick,sensitive and precise,and is suitable for the analysis of methylmalonic acid in the serum.
