江西省呼吸道合胞病毒G蛋白基因特征分析
Genetic characteristics of the G protein in RSV in Jiangxi province
【摘要】 目的了解江西省流行的呼吸道合胞病毒(RSV)的G蛋白基因特征。方法收集2005年-2007年流感样病例标本,进行RSV分离;对病毒G蛋白进行全基因序列测定,并与国际标准株进行比较,对G蛋白潜在的O-糖基化位点进行分析。结果从4291份标本中分离到6株RSV。各分离株与A、B型标准株的氨基酸同源性分别为85.6%~87.6%和75.9%~76.7%;其3’端基因高变区核苷酸序列与RSV GA2亚型的同源性最高。分离株在人群免疫选择压力位点111、132、226、262、274、290、297位氨基酸上发生了变异,与A2株比较,众多的潜在的O-糖基化位点发生了改变。结论 2005年-2007年江西省RSV流行株属于GA2亚型,在G基因的人群免疫选择压力位点和糖基化位点有变异。
【Abstract】 Objective To know the genetic characteristics of the G protein of prevalent RSV in Jiangxi province. Methods The throat swabs specimens were collected from influenza- like cases from 2005 to 2007. The whole region of the G gene of the isolates was sequenced and phylogenetic trees were constructed by comparison with RSV standard strains and reference strains.NetOglyc software( version 3. 1) was used to predict the potential o- glycosylated serine and threonine residues. Results Six RSV strains were isolated from 4291 throat swabs specimens. The identity of the G gene of the six isolated strains and that of the standard A and B type of RSV strain was 85. 6% ~ 87. 6% and 75. 9% ~ 76. 7% respectively. The sequence of the 3’hypervariable region of the G gene was highly identified with the GA2 subtype of RSV. The immunoselection pressure sites( 111,132,226,262,274,290,297aa) varied in the six isolates,and many potential O- glycosylation site had changed compared with GA2 subtype of RSV. Conclusion The isolates of RSV in Jiangxi province belong to the GA2 subtype,and variation was found in the immunoselection pressure site and glycosylation site of G gene.
