浒苔多糖的分离纯化和成分分析

【摘要】 目的研究浒苔多糖(EP)的分离纯化条件和结构特点。方法以福建浒苔为研究对象,采用二乙氨基乙基纤维素(DEAE纤维素)和Sephadex-100凝胶分离纯化浒苔多糖,用红外线光谱分析结构,用气相色谱分析单糖组成。结果浒苔多糖经DEAE纤维素分离得到4个组分,EP1、EP2、EP3和EP4,其中EP2在所回收的多糖中所占的比例最大,达到81%。EP2经Sephadex-100分离时,时呈现单一对称峰;红外线光谱分析显示,EP2为糖类化合物,且有硫酸酯结构;经气相色谱检测显示,EP2由鼠李糖、葡萄糖醛酸、阿拉伯糖、岩藻糖、甘露糖和半乳糖组成,各种单糖的比例为5.12∶1.32∶3.38∶1.62∶1∶1.03。结论 EP的主成分为EP2,EP2是一种含有硫酸酯的复合多糖。

【Abstract】 Objective To study the separation and purification condition and structure features of polysaccharide from Enteromorpha. Methods The Enteromorpha produced in Fujian province was taken as the research object. DEAE- cellulose and Sephadex- 100 were used to separate and purify the polysaccharide from Enteromorpha( EP). Infrared spectrum was used to analyze the structure of EP and gas chromatography was used to study the composition of monosaccharide of EP. Results Four components were separated from EP by DEAE- cellulose,they were EP- 1,EP- 2,EP- 3,EP- 4. EP2 accounted for the largest proportion of the recycled polysaccharide,reaching 81%. EP- 2 showed a single symmetric peak when separated by Sephadex- 100; Infrared spectrum showed that EP- 2 was a kind of carbohydrate with thiol esters. GS showed that EP- 2 was made up of rhamnose,glucuronic acid,arabinose,fucose,mannose and galactose,and the proportion of each monosaccharide was 5. 12∶ 1. 32∶ 3. 38∶ 1. 62∶ 1∶ 1. 03. Conclusion The major components of EP is EP- 2. EP- 2 is a kind of polysaccharide containing thiol esters.
