响应面分析法优化生姜中姜黄素的提取工艺研究
Optimization of the extraction of curcumin from ginger via response surface methodology
【摘要】 姜黄素是生姜中主要有效成分,具有多种生物活性,是生姜发挥药理作用的主要成分。本文运用超声波协同双水相的方法,利用响应面分析法对姜黄素的萃取条件进行优化研究。通过单因素实验考查不同有机溶剂体积、超声波时间及提取温度对生姜中姜黄素提取的影响。在单因素试验的基础上采用三因素三水平的响应面分析法,根据回归分析确定各工艺条件的影响因子,以上相提取率为响应值作响应面和等值线图。结果表明,姜黄素萃取的最佳工艺条件为:K2HPO46.0g、超声提取时间为30.2 min、乙醇体积9.86 m L、提取温度56.6℃,姜黄素的收率(Yield of Turmeric:Yt)为98.9%,有机相中提取率为0.054%。
【Abstract】 Curcumin is the main active component in ginger and has a variety of biological activity. It is also the main component of ginger’s pharmacological effects. The aim of this project was to evaluate the effect of dynamic factors upon the Curcumin from Ginger extraction. Response surface methodology was used to determine the optimum parameters for ultrasonic- assisted aqueous two- phase extraction of curcumin in ginger. Through the single factor experiment,different organic solvent volume,ultrasonic time and extraction temperature were tested on the extraction effect of curcumin. Based on the above result,three factors and three levels of response surface analysis was performed,and according to the regression analysis,the effects of process conditions factors,the top phase of extraction rate were chosen as response values for the response surface and contour map. The results showed that the optimum conditions of curcumin extraction were: fixed liquid adding amount of K2HPO46 g,ultrasonic extraction time 30. 2 min,volume of ethanol9. 86 m L,extraction temperature set 56. 6 ℃,Yt( yield of curcumin) 98. 9%. Under the above conditions,the organic phase of extraction rate was 0. 054%.
