高效液相色谱-柱后光化学衍生-荧光检测法测定强化食品中的叶酸

【摘要】 目的建立柱后光化学衍生-高效液相色谱法(HPLC)测定强化食品中叶酸的分析方法。方法采用SunFire C18(4.6 mm×250 mm,5μm)色谱柱,以乙腈-50 mmol/L磷酸二氢钾溶液(pH=5.0)为流动相梯度洗脱,叶酸在254 nm紫外光照射下进行柱后衍生,荧光检测器检测,激发和发射波长分别为344和428 nm。结果本方法的线性范围为0.01~2.0μg/ml,r=0.999 9;方法的定性检出限和定量检出限分别为0.01和0.03μg/g;加标回收率为90.0%~98.0%,RSD均小于5%。结论本方法简便、快速、准确、重现性好,适用于强化食品中叶酸含量的测定。

【Abstract】 Objective To establish a method for the determination of folic acid in fortified foods by high performance liquid chromatography( HPLC) with post-column photochemical derivatization. Methods Chromatographic separation was carried out using a SunFire C18column( 4. 6 mm × 250 mm,5 μm). The mobile phases was acetonitrile and50 mmol /L potassium dihydrogen phosphate solution( pH = 5. 0) with an optimized gradient elution. Post-column photochemical derivatization of folic acid was under UV-light irradiation at 254 nm. The fluorescence detection was performed with the excitation at 344 nm and the emission at 428 nm. Results The linear range was 0. 01-2. 0 μg /ml,r = 0. 999 9. The limit of detection( LOD) was 0. 01 μg /g and the limit of quantification( LOQ) was 0. 03 μg /g. The average recoveries were 90. 0%-98. 0% and the RSD of the method was less than 5%. Conclusion The method was simple,rapid,accurate,reproducible,and suitable for the determination of folic acid in fortified foods.
