中性粒细胞和单核细胞在脓毒症相关急性肺损伤聚集和来源差异的研究

【摘要】 目的在脓毒症模型的基础上,研究中性粒细胞与单核细胞在肺组织中聚集和来源的差异,探讨肺损伤的发生机制。方法按照10 mg/kg的剂量,给予大鼠腹腔注射细菌脂多糖建立全身脓毒症模型,通过流式细胞术检测模型鼠24 h内骨髓、脾脏、外周血、肺脏和肺泡灌洗液中中性粒细胞和单核细胞的动态变化。结果本研究通过动态观察发现与正常大鼠相比,脓毒症鼠肺脏中中性粒细胞和单核细胞数量增加,并且中性粒细胞聚集早于单核细胞,骨髓中两种细胞数量明显减少,然而脾脏仅有单核细胞减少。结论中性粒细胞在脓毒症相关肺损伤中发生聚集早于单核细胞,骨髓在脓毒症中作为中性粒细胞和单核细胞的来源,而脾脏仅作为单核细胞的来源。

【Abstract】 Objective To investigate the role of neutrophils and monocytes in lung injury and explore the mechanism of sepsis related lung injury. Methods The sepsis rat model was established by intraperitoneal injection of lipopolysaccharide of 10 mg/kg dosage. The dynamic changes of bone marrow, spleen, peripheral blood, lung and bronchoalveolar lavage fluid of model rat were detected by flow cytometry combined with absolute cell count in 24 hours. Results Compared with normal rat, the number of neutrophils and monocytes increased significantly in lung in sepsis model, and the increase of neutrophils was earlier than monocytes. Both two kinds of cells significantly reduced in bone marrow, whereas only monocytes were reduced in spleen. Conclusions Neutrophils appeared earlier than monocytes in sepsis related lung injury, and bone marrow was the source of neutrophils and monocytes, but spleen was only the source of monocytes in sepsis.
