益气解毒活络方对早期糖尿病肾病大鼠的防治作用
Protective and Therapy Effect of Compound Chinese Medicine Yiqi Jiedu Huoluo Fang in Rats with Diabetes Nephropathy
【摘要】 目的:观察益气解毒活络方对早期糖尿病肾病(diabetic nephropathy,DN)大鼠血流变和纤维蛋白原(Fib)及前列腺素E2(PGE2)的影响。方法:采用高脂高糖饲料喂养合并腹腔注射小剂量链尿佐菌素(streptozocin,STZ)方法建立糖尿病(DM)大鼠模型。以西药盐酸贝那普利作阳性对照,中药干预以益气解毒活络中药复方(12.78 g·kg-1)分别灌胃治疗8,10周。实验结束后,检测大鼠空腹血糖(FBG)、糖化血红蛋白(HbA1c)、血β2微球蛋白(β2-MG)及尿微量白蛋白(MALB);光镜观察肾组织形态结构变化(HE染色);血流变仪测定全血黏度(高切、中切、低切);凝血4项分析仪测定血浆Fib;酶联免疫吸附实验(Elisa法)检测血浆及肾组织PGE2的含量。结果:模型组大鼠FBG,HbA1c及血β2-MG、全血黏度、血浆Fib、尿MALB较正常组明显升高(P<0.05),PGE2含量升高(P<0.05);与模型组比较,各治疗组大鼠FBG,HbA1c及血β2-MG、全血黏度、血浆Fib、尿MALB显著降低(P<0.05),PGE2含量下降(P<0.05);模型组大鼠出现明显的肾脏形态学异常,各治疗组肾脏病变明显减轻。结论:DN的发病机制可能与血流变、Fib的异常及PGE2密切相关,益气解毒活络中药复方防治DN的发生发展可能是通过调节血流变、血浆Fib及纠正PGE2的异常实现的。
【Abstract】 Objective: To investigate the effects of compound Chinese medicine Yiqi Jiedu Huoluo fang on hemorheology and fibrinogen( Fib),prostaglandin E2( PGE2) level in rats with diabetes nephropathy( DN). Method: The rat model was established by feeding of HG and HF diet with peritoneal injection of streptozocin( STZ). Interfere with benazepril( 8 weeks) and herbs( 12. 78 g·kg-1,8,10 weeks). Fasting blood-glucose( FBG),haemoglobin A1c( HbA1c) and β2-microglobulin( β2-MG) and microalbuminuria( MALB),PGE2( Elisa) were tested. The renal pathological changes were evaluated by HE staining. Observed the changes of hemorheology and Fib, PGE2. Result: The level of FBG, HbA1c,β2-MG, hemorheology and Fib, MALB, PGE2in the model group were higher obvious( P < 0. 05),the other group were lowed. The renal pathological changes were obviously in the model group, and the therapy group were all alleviated. Conclusion: The pathological changes of kidney tissue have a realationship with hemorheology and Fib,PGE2,herbs of Yiqi Jiedu Huoluo fang in percent and cure to diabetic mellitus( DN) may by regulating the abnormal of hemorheology and Fib and PGE2.
