HPLC同时测定裸花紫珠中4种黄酮
Simultaneous Determination of Four Flavonoids in Callicarpa nudiflora by HPLC
【摘要】 目的:建立同时测定裸花紫珠药材中木犀草素-7-O-β-D-吡喃葡萄糖苷、木犀草素、5,4’-二羟基-3,7,3’-三甲氧基黄酮和5-羟基-3,7,3’,4’-四甲氧基黄酮含量的反相高效液相色谱法。方法:采用HPLC,Venusil XBP-C18色谱柱(4.6 mm×200mm,5μm),流动相甲醇-体积分数为0.1%磷酸水系统,梯度洗脱,流速1.0 mL·min﹣1,柱温室温,检测波长350 nm。结果:在上述色谱条件下测得木犀草素-7-O-β-D-吡喃葡萄糖苷、木犀草素、5,4’-二羟基-3,7,3’-三甲氧基黄酮和5-羟基-3,7,3’,4’-四甲氧基黄酮的质量浓度分别在0.000 8~0.016,0.001 6~0.032,0.004~0.08,0.002 4~0.048 g·L-1时与色谱峰面积之间线性关系良好;平均回收率为98.5%(RSD 1.7%),98.6%(RSD 1.6%),98.6%(RSD 1.4%),98.6%(RSD 1.7%)。结论:该方法精密度高,重复性好,可用于裸花紫珠药材中木犀草素-7-O-β-D-吡喃葡萄糖苷、木犀草素、5,4’-二羟基-3,7,3’-三甲氧基黄酮和5-羟基-3,7,3’,4’-四甲氧基黄酮的含量测定。
【Abstract】 Objective: To develop HPLC for the simultaneous determination of four flavonoids in Callicarpa nudiflora. Method: The separation was performed on a Venusil XBP-C 18 column( 4. 6 mm × 200 mm,5 μm),the analytes were separated efficiently using the mobile phase consisted of MeOH-H 2 O [w( H 3 PO 4) = 0. 1%]in a gradient program. The flow rate was maintained at 1. 0 mL·min﹣1. The detective wavelength was set at 350 nm and the column temperature was 25 ℃. Result: There were good linear relationships between the concentrations and the peak area of luteolin-7-O-β-D-glucopyranoside,luteolin,5,4’-dihydroxy-3,7,3’-trimethoxyflavone and 5-hydroxy-3,7,3’,4’-tetramethoxyflavone in the ranges of 0. 000 8-0. 016 g·L-1( r = 0. 999 1),0. 001-0. 032 g·L-1( r =0. 999 1),0. 004-0. 08 g·L-1( r =0. 999 2),0. 002 4-0. 048 g·L-1( r = 0. 999 1),respectively. The recoveries were found to be 98. 5% with RSD of 1. 7%( n = 9),98. 6% with RSD of 1. 6%( n = 9),98. 6% with RSD of 1. 4%( n = 9) and 98. 6% with RSD of 1. 7%( n = 9),respectively.Conclusion: The method is accurate and reproducible for determination of luteolin-7-O-β-D-glucopyranoside,luteolin,5,4’-dihydroxy-3,7,3’-trimethoxyflavone and 5-hydroxy-3,7,3’,4’-tetramethoxyflavone in C.nudiflora.
