【摘要】 目的调查云南省流行性乙型脑炎(乙脑)患者感染乙脑病毒(JEV)基因型状况。方法在云南省西双版纳州医院和大理州医院采集脑炎患者急性期血清标本20份和脑脊液标本11份,用RT-PCR法检测JEV核酸,阳性者进行JEV-PreM/C区基因序列测定,用核苷酸同源性和系统进化分析进行基因型鉴定。结果用RT-PCR法从11例脑炎患者脑脊液标本中检测到JEV核酸阳性3份(JB114、JB135和DLN24),而20例脑炎患者血清标本均为阴性。3株病毒的PreM/C区核苷酸序列同源性分析显示,JB114、JB135与JEV基因Ⅰ型代表株同源性高达98.3%和99.2%,而与Ⅱ、Ⅲ、Ⅳ型仅为82.5%~84.6%;DLN24与基因Ⅲ型代表株同源性为94.6%,而与基因Ⅰ、Ⅱ、Ⅳ型分别为85.0%、82.5%和80.4%。进化树分析显示,JB114和JB135位于基因Ⅰ型分支中,DLN24位于基因Ⅲ型分支上。结果表明,来自西双版纳的JB114和JB135病毒株属于基因Ⅰ型,来自大理的DLN24病毒株为基因Ⅲ型。结论云南省乙脑患者中分别存在基因Ⅰ型和Ⅲ型JEV感染的病例,表明云南省存在这两个基因型病毒的共同流行。更多还原

【Abstract】 Objective To investigate the genotype distribution of Japanese encephalitis virus(JEV)from encephalitis patients in Yunnan province,China.Methods Serum and cerebrospinal fluid(CSF)samples from encephalitis patients were collected in Yunnan province.RT-PCR was performed using JEV PreM/C specific primers.Positive specimens were sequenced for homology and phylogenetic analysis.Results Three out of 11 CSF samples(JB114,JB135 and DLN24)were positive for JEV nucleic acid while 20 serum samples were negative.JB114 and JB135showed higher homology in nucleic acid sequences with genotype I JEV(JEV/sw/Mie/40/2004)(98.3%and 99.2%,respectively);while the nucleotide homology of JB114,JB135 with genotypeⅡ,Ⅲand IV JEV strains were only 82.5%-84.6%.DLN24 and genotypeⅢJEV(Nakayama)showed higher nucleotide homology at 94.6%.The nucleotide homology of DLN24 with genotypeⅠ,Ⅱ,and Ⅳ JEV strains were 85.0%,82.5% and 80.4%,respectively.Phylogenetic analysis showed that JB114,JB135 strains from Xishuangbanna district of Yunnan province belong to genotype I,and DLN24 strain from Dali district of Yunnan province belongs to genotypeⅢ.Conclusions GenotypesⅠandⅢJEV infection of Japanese encephalitis were identified in Yunnan province of China.更多还原
