白细胞介素23与小鼠支气管哮喘相关性研究
Correlation Study of Interleukin-23 and Bronchial Asthma in Mouse
【摘要】 目的通过检测哮喘鼠血清中白细胞介素23(IL-23)在哮喘组和对照组中的水平及与IL-5,IFN-γ,IL-17之间的相关性,探讨其在哮喘病中的作用.方法将24只健康雌性BALB/C小鼠随机分为两组,卵白蛋白(OVA)致敏并激发的哮喘模型组(Asthma,简称A组)和正常对照组(Control,简称C组),每组12只,OVA致敏末次激发24 h后,各组小鼠眶静脉取血,计数支气管肺泡灌洗液(BALF)中白细胞总数并分类.ELISA法检测血清IgE,IL-5,IFN-γ,IL-17和IL-23的含量,取肺组织行组织病理学检测.结果 A组病理学改变、动物行为学改变、血清IgE水平增高均证明哮喘模型制备成功;IL-5显著增高,IFN-γ降低;A组小鼠血清中IL-23,IL-17水平高于C组,IL-23与IL-17含量成正相关关系(r=0.763,P<0.05),与IL-5,IFN-γ无相关性.结论 IL-23与哮喘发病密切相关,其机制可能是通过调节IL-17的产生促使哮喘发病;IL-23对IFN-γ和IL-5无明显调节作用.
【Abstract】 Objective To measure serum interleukin-23(IL-23) IL-5,IFN-γ and IL-17 levels of mice in asthma(A)and control(C) group,and analyze the correlations of IL-23 levels with IL-5,IFN-γ and IL-17 levels to explore the role of IL-23 in bronchial asthma. Method Twenty-four female BALB /C mice were randomly divided into two groups: mice in group A(asthma model) were sensitized and triggered by Ovalbumin(OVA),and those in group C(control group) were treated with PBS buffer. OVA at the 24th hour after the last challenge mice’s blood samples were obtained through the orbital vein. The white blood cells count and differential count in BALF(bronchoalveolar lavage fluid) were carried out. Serum IgE,IL-5,IFN-γ,IL-17 and IL-23 levels were measured by enzyme-linked immunoabsorbent assay(ELISA),and the lung tissues were removed for the pathological examination. Results Changes in pathology,behavior and serum IgE levels specific to asthma indicated that the establishment of asthma animal model was successful. The results showed the level of serum IL-5 increased and the level of IFN-γ decreased; levels of serum IL-23 and IL-5 in group A were higher than those in group C,and there was a positive correlation between the levels of serum IL-23 and IL-17(r = 0. 763,P<0. 05); but there was no correlation between IL-5 and IFN-γ. Conclusion IL-23 may be closely related to the onset of asthma,it can promote the asthma attack by regulating the production of IL-17; IL-23 has no significant regulatory effect on IFN-γ and IL-5.
