microRNA-椎间盘疾病治疗的新领域
microRAN- the new field of intervertebral disc diseases treatment
【摘要】 腰痛是临床常见病和多发病,而腰椎间盘退变(intervertebral disc degeneration,IVDD)是引发腰痛的重要原因之一。椎间盘发生退变时,最主要的病理改变是细胞外基质降解加速和细胞凋亡数目增加,导致椎间盘正常结构破坏。目前临床上对于IVDD的治疗效果仍不甚满意,而组织工程修复技术的兴起,尤其基因技术领域的开启为IVDD的治疗带来了新的曙光。MicroRNAs是一类内源性非编码小RNA,其常规作用方式是通过与靶基因的3’UTR结合抑制靶基因的翻译过程,且其多靶点的作用方式,方便了microRNAs的临床应用。MicroRNAs与IVDD的关系已有相关报道,但研究局限,尚需从多方面进行深入探索。
【Abstract】 Low back pain,as a common and frequently- occurring disease in clinical practice,is mainly caused by intervertebral disc degeneration( IVDD). The major pathological changes in degenerated disc include acceleration of extracellular matrix degradation and enhancement of cell apoptosis,which results in the destruction of normal disc structure. Currently,no effective clinical management for IVDD has been developed. A new dawn has been brought for the treatment of IVDD due to the development of tissue engineering technology,especially gene technology. MicroRNAs,a class of endogenous,short,non- coding RNA molecules,mainly function in suppressing the translation of target gene by binding to the 3` untranslated region( UTR) of the gene. Moreover, the characteristic of multiple functional sites of microRNAs greatly facilitate their clinical application. Although the relationship between microRNAs and IVDD has been reported by some previous work,a series of following studies wait to be carried out to reveal the precise regulation mechanisms of IVDD by microRNAs.
