15种竹叶中碳苷黄酮的含量研究

【摘要】 为找到富含竹叶黄酮的竹类资源.采用高效液相色谱-紫外分光光度法(HPLC-UV)同时测定刚竹属15种竹子中6种碳苷黄酮的含量,Eclipse XDB-C18色谱柱(4.6mm×250mm,5μm),流动相混合溶剂A(乙腈)和溶剂B(体积分数0.2%乙酸水)梯度洗脱条件:10%A(34min),10%~13%A(34~52min),13%~15%A(52~85min),15%~10%A(85~90min),流速1.0mL/min,检测波长330nm,柱温30℃,进样量20μL.结果表明:5种竹子:毛竹、淡竹、天目早竹、龟甲竹和斑竹富含碳苷黄酮类化合物,可作为碳苷黄酮的主要来源.

【Abstract】 To find more and new species of bamboo which are rich in bamboo leaf flavones,a strategy for analyzing flavone C-glycosides in the leaves of different cultivars of bamboo was developed.An HPLC-UV method was optimized and validated for determination of six flavone Cglycosides in bamboo leaves from 15cultivars of Phyllostachys.Chromatographic separations of these compounds were performed on an Ecilipse XDB-C18column(5μm,250mm×4.6mm i. d.).A gradient programme was used with the mobile phase,combining solvent A(acetonitrile) and solvent B(0.2%,acetic acid adjusted to pH 3.0)as follows:10% A(34min),10%~13% A(34~52min),13%~15% A(52~85min),15%~10% A(85~90min).The flow rate was 1.0 mL/min,the injection volume was 20μL and the column temperature was maintained at 30°C. Signal was monitored at 330nm with UV.Five cultivars(P.heterocycla,P.glauca,P. tianmuensis,P.pubescens and P.bambusoides)are rich in flavone C-glycosides.
