在体鸡胚听骨染色与解剖形态演变

【摘要】 目的建立正常鸡胚发育过程中听骨(耳小柱)解剖形态与特征的参照标准,同时探索在整体标本中清晰显示耳小柱的方法。方法发育第5-19天(E5-E19)鸡胚头部,经阿辛蓝软骨染色、甲醛水杨酸软组织透明后,显微镜下观察耳小柱及其周围软骨发育情况。结果耳小柱在E7出现,E8-E10呈分离的内、外侧2部分,E11耳小柱及周围软骨已具成熟期形态,E12形态更清晰,E13软组织内血管影出现,E14软骨开始钙化,E15后软骨形态已无明显改变,但软组织透明度降低、血管影增多。结论耳小柱均E7出现,E11时已具备成熟期形态。阿辛蓝软骨染色、甲醛水杨酸软组织透明法能在整体标本中清晰显示位置深在的耳小柱形态和结构。

【Abstract】 Objective Master normal morphology and establish a criterion of chick columella during its development,and find a method which can show the configuration of deep located columella in the whole mount. Method Collected E5-E19 chick embryos’ heads, fixed with 4%PFA, stained cartilages with Alcian Blue, cleared tissue by methyl salicylate, and ob-served columella and other cartilages around by anatomy microscope. Results Columella came out on E7, keeping as 2 sepa-rate parts during E8- E10, displayed matured structure on E11(also other cartilages around it), on E12, columella’s configu-ration was clearer, While blood vessel’s shade came out on E13 and calcification began on E14. After E15, there were no mor-phology changes found, but transparence of connective tissue decreased and blood vessel’s shade increased. Conclusions Columella came out on E7 and showed matured morphology on E11(also other cartilages around it), and it is a good method forshowing deep located columella in the whole mount to stain cartilages by Alcian Blue and clear tissue with methyl salicylate.
