【摘要】 目的 探讨低分子量蛋白酶体2(LMP-2)/LMP-7基因多态性与大兴安岭地区哈萨克族人群食管鳞癌的关系。方法 运用两步法聚合酶链反应技术及免疫组化方法研究LMP2/LMP7基因在大兴安岭地区汉族与哈萨克族食管鳞癌患者血液中的表达情况,并与该地区健康人群对比。结果 300例哈萨克族食管鳞癌患者中188例(62.7%)LMP2/LMP7基因表达呈阳性,其中LMP-2检测出5种分型,H型占9.6%(18/188),R型占23.9%(45/188),H/H型占2.7%(5/188),H/R型占11.2%(21/188),R/R型占3.2%(6/188);LMP-7检测出5种分型,Q型占18.6%(35/188),K型占10.6%(20/188),Q/Q型占4.8%(9/188),Q/K型占4.3%(8/188),K/K型占1 1.2%(21/188)。300例汉族食管鳞癌患者中仅2例(0.7%)阳性,其中LMP-2检测出的5种分型比率分别是H型占10.0%(2/20),R型占15.0%(3/20),H/H型占5.0%(1/20),H/R型占0.0%(0/2),R/R型占20.0%(4/20);LMP-7的5种分型中Q型占10.0%(2/20),K型占3.5%(7/200),Q/Q型占25.0%(5/20),Q/K型占5.0%(1/20),K/K型占6.5%(13/200)。对照组(汉族和哈萨克族人各300例)检测结果为阴性。结论 LMP-2/LMP-7基因高表达可能与哈萨克族食管鳞癌密切相关,可以作为判断病情和预后评价的指标。更多还原

【Abstract】 Objective To investigate the polymorphism of low molecular weight protease(LMP2/LMP7)and its association with susceptibility to esophageal squalors cell carcinoma(ESCC) in Kazakh in Daxinanling.Methods LMP2/LMP7 genotypes were detected by RT-PCR and immunohistochemical method in Kazakh ESCC and The not-cancer patients in the same area.Results Totally 188 cases of patients with positive ESCC in 300 patients were detected in Kazakh(62.7%).Five kinds of genotypes of LMP-2 were found.H type was 18 cases(9.6%);R type was 45 cases(23.9%);H/H type was 5 cases(2.7%);H/R type was 21 cases(11.2%);R/R type was 6 cases(3.2%),Five kinds of genotypes of LMP-7 were found.Q type was 35 cases(18.6%);K type was 20 cases(10.6%);Q/Q type was 9 cases(4.8%);Q/K type was 8 cases(4.3%);K/K type was 21cases(11.2%),About 2 cases of patients with positive in 300 patients with ESCC were detected in the blood in Han ethnic(0.7%).Five kinds of genotypes of LMP-2 were found.H type was 0.2 cases(10.0%);R type was0.3 cases(15.0%);H/H type was 0.1 cases(5.0%);H/R type was 0 cases(0.0%);R/R type was 0.4 cases(20.0%),Five kinds of genotypes of LMP-7 were found.Q type was 0.2 cases(10.0%);K type was 0.07cases(3.5%);Q/Q type was 0.5 cases(25.0%);Q/K type was 0.1 cases(5.0%);K/K type was 0.13 cases(6.5%).The control lest results were negativ(P<0.05).Conclusion LMP2 / LMP7 gene high expression may be closely related with the squamous cell esophageal cancer.更多还原
