日粮不同能量水平对乌金猪肌内脂肪沉积相关基因表达的影响
Effect of Dietary Energy Levels on Gene Expression Related With Intramuscular Fat Deposition in Muscle Tissue of Wujin Pigs
【摘要】 本试验旨在研究日粮能量水平对乌金猪不同生长阶段肌内脂肪沉积候选基因表达水平的影响。选取体重约15 kg的乌金猪54头,随机分成3组,每组3个重复,每个重复6头,在15~30、30~60、60~100 kg体重阶段分别饲喂不同能量水平日粮,日粮水平为低能量水平(消化能11.84MJ/kg)、中能量水平(消化能12.97 MJ/kg)和高能量水平(消化能14.18 MJ/kg),在体重30、60、100 kg时分批屠宰取背最长肌测定肌内脂肪含量,通过Real-tine PCR检测肌内脂肪沉积候选基因mRNA表达水平。结果表明:高能量组乌金猪背最长肌肌内脂肪含量显著高于中、低能量组(P<0.05);乌金猪背最长肌中LEPR、RBP4、MC4R基因表达水平均随能量水平的升高而升高,高能量组各基因的表达水平显著高于中、低能量组(P<0.05)。总之,高能量饲粮显著提高乌金猪肌内脂肪含量以及显著促进乌金猪背最长肌LEPR、RBP4、MC4R基因的表达水平。
【Abstract】 The aim of the study was to investigate the effects of dietary energy levels on gene expression related intramuscular fat deposition in muscle tissue of Wujin pigs.Fifty-four Wujin pigs with the body weight of 15 kg were randomly assigned into 3 groups which was imposed on three different energy levels.There were eighteen pigs in each group with three replicates containing six pigs for each replicate.The dietary energy levels were high energy group(DE,11.84 MJ /kg),medium energy group(DE,12.97MJ/kg) and low energy group(DE,14.18 MJ /kg).At the body weights of 30,60 and 100 kg,pigs were slaughtered and longissimus muscle sample were taken for analyzing the expression levels of genes related intramuscular fat deposition by Real-time PCR.The results showed that the content of intramuscular fat of pigs in high dietary energy group was higher significandy than that in middle and low dietary energy group(P<0.05).The gene expression levels of LEPR,RBP4,MC4 R genes in longissimus muscle of Wujin pigs was increased with dietary energy levels’ increasing,and the expression levels of all genes in high dietary energy group was significantly higher than that in middle and low dietary energy group(P<0.05).Therefore,the diet with high energy can obviously improve the content of intramuscular fat and increase the expression levels of LEPR,RBP4,MC4 R genes in longissimus muscle of Wujin pigs.
