应用16 S rRNA基因序列分析快速鉴定临床标本中的革兰氏阳性杆菌

【摘要】 目的探索一种快速鉴定临床标本中革兰氏阳性杆菌的方法。方法利用PCR技术扩增待检菌株的16 S rRNA基因序列,通过分析待检菌株的16 S rRNA基因序列对其进行鉴定。结果 5株待检菌株的16 S rRNA基因序列均成功扩增,其中4株的16 S rRNA基因序列与基因库中已注册的核酸序列相似率达99.9%以上,将其鉴定到种的水平,1株的16 S rRNA基因序列与基因库中雷弗森菌属的核酸序列相似率为97.09%,将其鉴定为雷弗森菌属。结论应用16 S rRNA基因序列分析可快速、准确地鉴定临床标本中的革兰氏阳性杆菌。

【Abstract】 Objective To explore a rapid method for identifing Gram-positive bacilli in clinical specimens. Methods 16 S rRNA gene sequence of strains to be detected was amplificated by PCR technology,and these strains were identified using 16 S rRNA gene sequence analysis. Results 16 S rRNA gene sequence of 5 strains to be detected were amplificated successfully,among them,the similar rate of between 16 S rRNA gene sequence in 4 strains and the nucleic acid sequence registered of gene library was more than 99.9%,and these were identified to the species level,the similar rate of between 16 S rRNA gene sequence in 1 strain and the nucleic acid sequence of Refson sp. of gene library was 97.09%,and this was identified as Refson sp. Conclusion The Gram-positive bacilli in clinical specimens could be identified rapidly and accurately using 16 S rRNA gene sequence analysis.
