五鹤续断抗小鼠衰老的作用及其机制研究

【摘要】 目的:研究五鹤续断(WHDA)注射液对D-半乳糖所致小鼠衰老模型的抗衰老作用及其机制。方法:昆明种小鼠(雌雄各半)48只,随机分为对照组、模型组、阳性WHDA组、7.2 g/kgWHDA组、3.6 g/kgWHDA组及1.8 g/kgWHDA组,每组8只。腹腔注射D-半乳糖造模,Morris水迷宫测量小鼠学习和记忆能力。检测各组小鼠皮肤羟脯氨酸、脑组织MDA、LP、SOD及GSH-Px含量,采用双抗体夹心ELISA法检测小鼠血清中IL-2及IL-6含量。结果:Morris水迷宫实验结果表明WHDA各组潜伏期与模型组相比较明显减少(P<0.05),模型组小鼠穿越平台的次数比其它各组小鼠少(P<0.05)。脑组织氧化性指标(SOD、MAD、LP及GSH-Px)检测,对照组及WHDA各组MAD与LP含量低于模型组(P<0.05)。对照组及WHDA各组SOD与GSH-Px的活性明显高于模型组(P<0.05)。注射过D-半乳糖小鼠皮肤羟脯氨酸含量都明显低于对照组(P<0.05),WHDA组小鼠皮肤中羟脯氨酸的含量明显高于模型组(P<0.05)。WHDA组小鼠血清中IL-2、IL-6含量显著高于对照组和模型组(P<0.05),模型组小鼠血清中的IL-2、IL-6比对照组要低(P<0.05)。结论:WHDA能改善D-半乳糖所致的衰老小鼠的学习记忆能力,消除其体内自由基,增强机体免疫能力,具有较好的抗衰老作用。

【Abstract】 Objective:To study the effect of Wu-He Dipsacus asper(WHDA),Traditional Chinese Medicine,injection on mice-aging model induced by D-galactose.Methods:Forty-eightKunmingmice(24 male and 24 female) were randomly divided into control group,model group,positive control group,7.2 g/kg WHDAgroup,3.6 g/kg WHDAgroup and 1.8 g/kg WHDAgroupwith eight in each group.The model was induced through injecting D-galactose into peritoneal cavity and Morris water maze was used to detectthe learning and cognitive ability of mice.The sk in hydroxyproline,bra in tissue malondialdehyde(MDA),lipofuscin(LP),superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)levels of mice were detected;the IL-2 and IL-6 levels in serum of mice were detected by using double antibody sand wich ELISA method.Results:Each WHDA group was significantly reduced in latency period compared with the model group during Morris water maze test(P< 0.05)and the number of mice in model group through the platform was less than othermice in each group(P< 0.05).The levels of MAD and LP of the control group and each WHDAgroup were less than model group in the detection of heart,bra in tissue oxidation index(SOD,MAD,LP and GSH-Px,P< 0.05).The activity of SOD and GSH-Px in the control group and each WHDA group was significantly higher than that in the model group(P< 0.05).The sk in hydroxyproline contentof mice which had been injected with D-galactose was significantly lower than that in the control group(P< 0.05)and the sk in hydroxyproline content of mice of WHDA group was significantly higher than that in the model group(P< 0.05).The IL-2,IL-6 levels in serum of mice in WHDAgroup were significantly higher than those in the control group and the model group(P< 0.05)and the IL-2,IL-6 levels in serum of mice in the model group were lower than those in the control group(P< 0.05).Conclusion:The effective constituents ofWHDAhave a variety of biological activitywhich can have a good effect on anti-aging by different ways,improving learning and memory function,eliminating free radicals antioxidant,and enhancing the body immunity and other aspects.
