肠炎沙门菌fimA、flhD基因启动子克隆及其LacZ报告质粒的构建
Construction of LacZ Reporter Plasmids with fimA and flhD Promoter in Salmonella enteritidis
【摘要】 肠炎沙门菌是引起人食源性疾病的主要人兽共患病原菌之一,其感染机制与细菌染色体上特定的毒力岛以及毒力质粒上的某些毒力基因有关。基于对肠炎沙门菌毒力基因的前期研究,以毒力因子SEF21菌毛编码基因fim A和鞭毛调控基因flh D为研究对象,通过PCR从肠炎沙门菌50336中扩增靶基因的上游启动子序列Pfim A和Pflh D,克隆到质粒p RS551中构建lac Z报告质粒,并将其转化入肠炎沙门菌50336中,再通过β-半乳糖苷酶试验检测其活性,为进一步利用Lac Z报告系统定量研究肠炎沙门菌内SEF21菌毛和鞭毛的毒力表达奠定基础平台。
【Abstract】 Salmonella enteritidis is a major pathogen of zoonoses,causes foodborne disease in human,the infection pathogenesis of which is related to specific virulence island on chromosome and some virulence genes on the virulence plasmid. Based on our previous study about specific virulence genes of S.enteritis,we targeted the important virulence factors,fim A(encoding the major subunit of SEF21 fimbriae)and flh D(encoding the flagellar regulator),and amplified the promoter Pfim Aand Pflh Dfrom S.enteritis strain 50336 with PCR,then cloned the promoters into plasmid p RS551 to construct the Lac Z reporter plasmid,respectively. β-galactosidase assays were performed to test the activity of the constructed reporter plasmids introduced into strain 50336. This study was providing the experimental basis platform for quantifying the expression of SEF21 fimbriae and flagella by Lac Z reporter system in Salmonella enteritidis.
