吉林省2012年柯萨奇病毒A组16型基因亲缘关系及氨基酸位点变异分析
Phylogenetic Relationship and Amino Acid Sequence Variation of Coxsackie Virus Group A Type 16 Strains in Jilin Province in 2012
【摘要】 目的了解吉林省2012年手足口病(Hand,Foot and Mouth Disease;HFMD)来源的柯萨奇病毒A组16型(Coxsackie Virus Group A Type 16,CVA16)基因亲缘关系及氨基酸位点的变异情况。方法收集吉林省2012年73例HFMD临床标本,病毒分离后对其中22株的VP1编码区部分核苷酸进行序列测定和分析。结果 22株CVA16中,12株为B1a基因亚型,10株为B1b基因亚型,12株B1a基因亚型氨基酸同源性为95.0%~97.8%,10株B1b基因亚型氨基酸同源性为94.3%~97.0%;有8株在第23位氨基酸位点发生了亮氨酸(Leucine,L)→蛋氨酸(Methionine,M)的变异,其余14株在该氨基酸位点均为L。结论 CVA16为吉林省HFMD的主要病原体之一,毒株间的同源性较高,均属于B基因群,在系统进化树上分别属于两个进化分支。
【Abstract】 Objective To determine phylogenetic relationship and amino acid sequence variation of coxsackie virus group A type 16( CVA16),the pathogen of hand,foot,and mouth disease( HFMD),in Jilin Province in 2012. Methods CVA16 strains were isolated from 73 patients with HFMD in Jilin province in 2012. VP1 gene fragments were sequenced and analyzed from 22 strains. Results Among the 22 strains,12 were B1 a sub-genotype; the rest were B1 b sub-genotype. Among the 12 B1 a strains,amino acid homology ranged from95. 0% to 97. 8%; among the other 10 B1 b strains,amino acid homology ranged from 94. 3% to 97. 0%. In 8strains,the amino acids at site number 23,which is normally leucine( L),mutated to methionine( M); the other 14 strains all had leucine at site number 23. Conclusion CVA16 strain is one of the most important pathogens causing HFMD in Jilin. The phylogenetic relationship among the strains was very close. All CVA16 strains from this study belonged to gene group B,and were from 2 phylogenetic branches.
