东港市2014年甲型肝炎病毒流行株基因分型研究

【摘要】 目的了解辽宁省东港市2014年甲型肝炎(甲肝)病毒(Hepatitis A Virus,HAV)流行株基因型特征,为HAV溯源研究奠定基础。方法收集东港市部分甲肝病人血清标本,HAV结构-非结构区基因VP1-2A引物,经核酸提取,逆转录-聚合酶链反应(Reverse Transcription-Polymerase Chain Reaction,RT-PCR),序列测定,对HAV基因进行序列同源性及亲缘关系分析。结果本次检测到的东港市HAV都属于1A亚型,核苷酸序列同源性为98.1%~100%,与中国河北省、日本及韩国的流行株VP1-2A编码区基因有相同序列或同源性较高。结论东港市有多株HAV存在,流行可能有多个传染链,在人群免疫水平较低时,引起甲肝流行。

【Abstract】 Objective To study the genetic characteristics of wild type hepatitis A viruses( HAV) that circulated in Donggang,Liaoning province of China in 2014. Methods VP1-2A coding region genes,which were isolated from serum samples obtained in 2014 from HAV patients in Donggang,Liaoning province,China,were amplified by Reverse Transcription-Polymerase Chain Reaction( RT-PCR) and then sequenced. The HAV gene sequence was analyzed for homology and phylogeny. Results The HAV strains were all sub-genotype 1A,and nucleotide homology was from 98. 1% to 100%. The VP1-2A coding region nucleotide sequences of Donggang strains had the same sequence,or showed strong homology,with the nucleotide sequence isolated in Hebei,China,Japan,and South Korea. Conclusion There were several HAV strains transmitted with different pathways in Donggang. HAV outbreaks will occur if population immunity is insufficient.
