钠葡萄糖共转运载体1在卵巢癌中的表达及作用研究

【摘要】 目的:探讨钠葡萄糖共转运载体1(SGLT1)在卵巢癌中的表达及其作用。方法:收集卵巢癌及癌旁正常组织,Western blot及PCR检测SGLT1蛋白及mRNA水平;培养类癌细胞系(carcinoid cancer)及卵巢癌细胞系(OVCAR3,SKOV3和A2780),WB及PCR检测SGLT1蛋白及mRNA水平;构建慢病毒小分子干扰SGLT1载体并筛选稳定细胞株,甲基偶氮唑蓝比色法(MTT)及BrdU掺入比色法检测细胞增殖情况,WB检测细胞周期相关蛋白的表达水平。结果:SGLT1在卵巢癌组织中表达上调,在卵巢癌细胞系OVCAR3中表达明显上调;慢病毒载体介导干扰SGLT1后,OVCAR3细胞P21及P27表达增加,p-Rb水平下调,OVCAR3细胞增殖被抑制。结论:SGLT1在卵巢癌中的表达上调,敲除SGLT1抑制OVCAR3细胞增殖。

【Abstract】 Objective: To explore the expression and effect of sodium-glucose transporter 1( SGLT1) in ovarian cancer. Methods: The ovarian cancer tissue and normal ovarian tissue adjacent to ovarian cancer were collected,Western blot and PCR were used to detect the levels of SGLT1 protein and mRNA; carcinoid cancer cell line and ovarian cancer cell lines( OVCAR3,SKOV3 and A2780) were cultured; lentiviral small- molecular interfering vector was established and stable cell strains were screened,MTT method and BrdU incorporation assay were used to detect cell proliferation,Western blot was used to detect the expression level of cell cycle- related proteins. Results: SGLT1 upregulated in ovarian cancer tissue,especially in ovarian cancer OVCAR3 cells; lentiviral vector- mediated interference of SGLT1 inhibited the growth and proliferation of OVCAR3 cells,increased the levels of P21 and P27 and reduced p- Rb. Conclusion:SGLT1 upregulates in human ovarian cancer; knockdown of SGLT1 inhibits the growth and proliferation of OVCAR3 cells.
